In order to reveal the physiological and protein basis on the fertility conversion of Photo-thermo-sensitive genic male sterility rice, the test was selected thermo sensitive genic male sterile rice Zhu1S,Zhun S,Annong81OS and Peiai64S as the materials, in the IV stage of panicle initiation with breeding temperature20.5℃fertility fluctuating temperatures22.5℃cold water continuous processing for7d, and using infertility temperature (average of daily temperature30.4℃) as the control, and according to processing time lh,6h,12h,24h,48h,96h,168h to sampling interval, study the regularity on physiological and biochemical dynamic variation in the young panicles and changes in pollen fertility, in the same time studying on the young panicle proteins of Zhu1S and Annong810S with proteomic technology,and the results were as follows:1. The pollen dyeable rate in four male sterile lines in different temperatures overall performed that20.5℃>22.5℃>30.4℃. And the longer the low temperature treatment was, the higher pollen dyeable rate in four male sterile line were improved. And Sensitivity to temperature performed that Annong810S> Zhun-S> Peiai64S> Zhu1S.And the selfing rate results in the four male sterile lines were consistent with the changes results of dyeable pollen rate.2. The young panicle of soluble protein, soluble sugar and proline content in four male sterile lines overall performed that20.5℃>22.5℃>30.4℃.During the conditions of20.5℃and22.5℃, the soluble protein, soluble sugar content and proline content in young panicle showed the same trend, that were rising for the first, after falling or flat rising and then quickly rising. And the SOD and CAT activity in the three kinds of temperature conditions were showed overall20.5℃>22.5℃>30.4℃,and three enzyme activities of dynamics changes were rose first and then decreased. MDA content overall performed to be30.4℃>22.5℃>20.5℃.In the three kinds of temperatures,the accumulation of young panicle soluble protein, soluble sugar and proline content and SOD activity, CAT activity with male sterile lines sterile turned to fertility showing a positive correlation,and POD activity and MDA content to fertility conversion was negatively correlated.3. Zhu1S and Zhun-S of young spike protein of differential expression,the results were obtained1100repeat proteins, the abundance of130protein spots differentially expressed, and Zhu1S had69differentially expressed proteins, Zhun-S had61differentially expressed proteins,successfully identified108differentially expressed proteins. And in these differentially expressed proteins involved all16kinds of metabolic pathways in the fertility transformation proces, mainly metabolic physiological processess were redox balance capacity, protein synthesis, amino acid metabolism, transcriptional regulation, cell cycle. Raised and lowered regularity of these metabolic pathways and physiological and biochemical of studying enzyme system change were consistent.4. The identification of differentially expressed proteins, most closely proteins to fertility conversion were phosphofructokinase,the26S proteasome,the uridine diphosphate Pyrophosphorylase and heat shock protein.
