The main composition of high complexation of povidone iodine solution is a high molecular bio-organic iodine which exists in the form of a low toxicity iodine complex. Appropriate concentration of iodine can kill the bacteria and viruses in the semen while not harm the sperm, effectively prevent the vertical transmission of bacterial and viral infectious diseases, greatly improve the overall quality of the semen. Due to no pollution to environment, it provides the high-risk farming a security barrier. In this breeding environment with many rampant diseases, the emergence of high complexation of povidone iodine solution can effectively prevents the boars’ disease inheriting to the next generation and provides a strong safeguards for the eiminating of the infectious disease of swine in the entire country. This issue confirms the killing effect of high complexation of povidone iodine solution to common clinical microorganisms through its sterilization virus test in vitro to common bacteria and viruses, and then discuss the applicable and feasibility of the application to the protection of livestock and poultry semen, provide a theoretical basis of its rational use in veterinary medicine production of semen normal temperature preservation.1. High complexation of povidone iodine solution vitro bactericidal testIn order to evaluate the vitro bactericidal activity of high complexation of povidone iodine solution, we studied the role of three dilution1:50,1:100,1:200of the test solution in the killing of enterobacteriaceae, pasteurella branch and gram-positive cocci by the suspension quantitative germicidal test under laboratory conditions, examining the average rate of sterilization and the killing of three dilution in the different time. the results showed that0.03mol·L-1phosphate buffer(0.1%lecithin+1.0%tween80+0.5%sodium thiosulfate)as a neutralizing agent can effectively neutralized the residual effects of1:50dilution of high complexation of povidone iodine solution.under laboratory conditions; the 1:50dilution of high complexation of povidone iodine solution as the sample solution’s5min sterilization rate can reach100%;1:100dilution of high complexation of povidone iodine solution for the sample solution’s of different strains of E. coli and Salmonella10min average sterilization rate is more than99%, the test average sterilization rate of gram-positive cocci were greater than99.9%;1:200dilution of high complexation of povidone iodine solution bactericidal effect is not ideal. Above tests showed that less than100times dilution of high complexation of povidone iodine solution has a good sterilization effect.2. The study of vitro anti-virus efficacy of high complexation of povidone iodine solution on PRRSV and TGEVThe aim of this research is to study the toxic effects of high complexation of povidone iodine solution and povidone iodine on Marc-145and PK-15. Studying the effect of high complexation of povidone iodine solution and povidone iodine on PRRSV and TGEV’s proliferation by MTT assay. The blocking, inhibition, direct killing PRRSV and TGEV test were done at the same time. Result:The maximum non-toxic concentration of high complexation of povidone iodine solution on Marc-145and PK-15are312.5μg·mL-1and156.25μg·mL-1. the maximum non-toxic concentration of povidone iodine on Marc-145and PK-15are2.44μg·mL-1and1.22μg·mL-1Below the safe concentration they will not cause significant damage and toxic effects on cells. The result of blocking test:At the safe concentration (1.22~312.50μg·mL-1), high complexation of povidone iodine solution has a highly significant effect on blocking the PRRSV (P<0.01). At the safe concentration (1.22~156.25μg·mL-1), high complexation of povidone iodine solution has a highly significant effect on blocking the TGEV (P<0.01). The result of inhibition test:at the safe concentration (2.44~312.50μg·mL-1), high complexation of povidone iodine solution has a highly significant inhibition effect on the proliferation of the PRRSV on cell Marc-145(P<0.01). At the safe concentration (4.88~156.25μg·mL-1), high complexation of povidone iodine solution has a highly significant inhibition effect on the proliferation of the TGEV on cell PK-15(P<0.01). The effect of direct killing test:at the concentration of0.15~312.50μg·mL-1, high complexation of povidone iodine solution has a highly significant direct killing effect on the proliferation of the PRRSV(P<0.01). At the safe concentration (19.53~156.25μg·mL-1), high complexation of povidone iodine solution has a highly significant direct killing effect on the proliferation of the TGEV(P<0.01). The result:At the safe concentration, high complexation of povidone iodine solution has a significant direct killing effect on virus. This experiment can provide a reference for using the high complexation of povidone iodine solution as a semen protection agent to prevent vertical transmission of semen disease from the source.3. The effect of high complexation of povidone iodine solution and povidone iodine on boar semen quality, survival time and survival indexThis experiment by studying the high complexation of povidone iodine solution and povidone iodine three dilution degrees (1:50,1:100,1:200) using1h after the effect of killing bacteria in pig semen, inspected the two kinds of solution of the effect of killing bacteria in pig semen; Of boar semen quality, indices such as survival time and survival index determination, study its impact on sperm quality, explore the feasibility of its application in livestock semen protection. Results show:1:100diluted high complexation of povidone iodine solution function can effectively kill most of the bacteria in pigs semen after1hours; The dose of17℃keep pigs had no effect on sperm quality of conventional indicators, sperm survival time can be effectively reached90h, overall survival time of up to300h, the highest survival index is105.3, the results were better than povidone iodine contrast group,1:50,1:200, diluted semen dilution protection effect is better than1:100. Conclusion:the comprehensive consideration of various factors in high dose of1:100diluted high complexation of povidone iodine solution can effectively kill the bacteria from the swine semen, the pig semen at room temperature preservation effect is good.4.
