Datura stramonium L. are important medicinal plants.Recently, with the pharmacological effects of scopolamine in-depth research and extensive application, there are many researches about it. By stimulating the rapid accumulation of elicitor hairy roots of secondary metabolites is one of the hotspot.In this research,the Agrobacterium Rhizogenes C58C1and leaves of Datura stramonium L. were used as materials.The leaves of Datura stramonium L. were infected by the agrobacterium,then the hairy root was produced after co-culture,sterilization and expand training,the dynamic changes of the hairy root would be detected.Then, research methyl jasmonate on Datura stramonium L. hairy roots growth, adversity physiological characteristics and the influence of the main content of secondary metabolites.The main results were as follows:1.Use Agrobacterium Rhizogenes C58C1infect Datura stramonium L. leaves, After co-culture5d, light culture around10d, induce hairy roots. Hairy roots of Datura stramonium L. in1/2MS liquid medium grew well, growing faster. Round lumps of hairy root formation, The middle of the old roots, darker.Outward light color, is the new root. This is related to the round-bottom flask and regularity circumference rotating shaker.2. According to experiments,the hairy root which was cultured in the liquid medium has several features:the growth of Datura stramonium L. hairy root has delaied periods (0-18d),rapid growth periods(18-27d) and moderate growth periods(27-30d). In Datura stramonium L. hairy roots,scopolamine and hyoscyamine production increased with the increase of hairy root culture time. When Datura stramonium L. hairy roots culture in about27d, synthesis of scopolamine and hyoscyamine reach a maximum, moderate growth periods of growth hairy roots, scopolamine and hyoscyamine synthetic slowed down.The experimental results show that the peak of Datura stramonium L. hairy roots growth is consistent with the peak of alkaloid synthesis are left in the27d.According to the growth and the alkaloid synthesis of hairy roots,the proper time to add elicitors should be the18th day.3.The experimental results show that methyl jasmonate can inhibit the growth of Datura stramonium L. hairy roots. Different concentrations of MJ were induced, hairy roots apical appear yellowing and browning. With200μ mol· L-1MJ to induced Datura stramonium L. hairy roots, fresh weight and dry weight of proliferation multiples are higher than other treatment,growth peak ahead of time3d. Hairy roots fresh weight and dry weight after treatment were significantly lower than the control without adding MJ. After treated by MJ, the contents of O2-, soluble protein, MDA, pro of Datura stramonium L. plant hairy roots increased in different degrees or first increase and then decrease. The contents of SOD activity and POD activity first increased and then decreased, stabilized. The contents of CAT activity showed decreased first and then increased and then decreased,or tends to be stable. Three kinds of protective enzyme activity changes associated with induction of concentration and induction time.The results illustrate, MJ affects Datura stramonium L. hairy roots oxidase activity.4.The experimental results show that,in Datura stramonium L. hairy roots,MJ could significantly promote the accumulation of scopolamine composition and release into the culture medium.Can significantly promote the hyoscyamine composition accumulation and release to medium, after treatmented3d and6d.
