This study chooses the reactivity of goatpox weak poison vaccine on goats as the model,to research the influence of genetic heterozygosity on goat humoral immune response, aimed to found the relationship between Black-boned goat individual gene heterozygosity and humoral immune response, and to explore the genetic effect of INH beta A gene on humoral immune response, we hope to provide beneficial reference for goat disease-resistant breeding in the future.We selected186healthy Black-boned goat to inject goatpox weak poison vaccine.The way of vaccination is to choose the goat tail medial intradermal inoculation,the dose is0.5ml per goat.To determine serum antibody concentration before immunization and on28days after immunization by used the goatpox antibody enzyme-linked immunoassay kits (the detection range:3.5ng/L-120ng/L). To extract the total genomic DNA from all goat samples, and to chooses13SNP loci from PGR、 GnRHR、FSHbβ、Mc1R、Agouti、INHβA、Myf5、AA-NAT、IGF-1、NPY-Y1R genes to do genotyping by using PCR-SSCP and PCR-RFLP method. PGR、GnRHR, FSHβ、LHβ、Mc1R、Agouti、Myf5、AA-NAT、IGF-1、NPY-Y1R gene loci were choosed to calculate individual genetic heterozygosity, then to analysis and comparise antibody levels of goatpox weak poison vaccine among different levels of heterozygosity; And to analysis goatpox virus antibody concentration, gene additive effect and dominant effect among different genotypes of INH beta A.The results showed that antibody concentration of the goatpox virus in serum is16.54ng/L and28.27ng/L before immunization and on28days after immunization respectively,The latter’s antibody concentration is significantly higher than the former one(p<0.01). Antibody concentration before immunization and on28days after immunization didn’t show significant difference among different genotypes of the12SNP locus, similarly, antibody concentration increment didn’t also show significant difference among different genotypes of the12SNP locus, so to choose the12SNP neutral locus to calculate individual genes heterozygosity.By analysis the correlation between antibody concentration increment and individual genes heterozygosity,we found that antibody concentration increment was reduction with individual genes heterozygosity rise, this showed weak the negative correlation. At that, it is a weak negative correlation between individual genes heterozygosity and the body’s immune response; In the study, INH beta A gene existed three kinds of genotype for GG, GH and HH, before immunization and on28days after immunization, goatpox virus antibody concentration among the three genotypes didn’t show significant differences (p>0.05), however, antibody concentration increment among the three genotypes showed the significant differences, GG type> GH type> HH type, antibody concentration increment of GG type is twice as tall as HH type one, the difference is very significant(p<0.01),So we concluded that INH beta A gene and reactive of goatpox virus vaccine has a significant genetic effect, however, if the gene can as a genetic markers or master genes that reflected animal humoral immune response ability, we need to do further research.
