Functional genomics is currently focus of plant genomics researches. Recently, the publication of wheat genome sequence accelerates the process of gene isolation and functional study. Using several molecular techniques, the present study analyzed the function of a resistance gene analog RGA2185, which was isolated from einkorn accession TA2033resistant to powdery mildew.mRNA epression assays showed that the expression level of RGA2185was induced by Bgt2, and peaked at72h after inoculation. There was no obvious phenotype on inoculated leaves of VIGS-silenced plants. At the same time, RGA2185driven by two different promoters was transformed into a wheat cultivar susceptible to powdery mildew by particle bombardment. Each transformation produced one positive To transgenic plant, and their T1generations showed different level of resistance to powdery mildew. Moreover, these transgenic lines also showed different expression level of RGA2185. Therefore, expression of RGA2185in wheat could improve resistance to powdery mildew.
