Waterfowl are the natural host of avian influenza virus and almost all subtypes currently known can be isolated from them so far. Early study revealed that waterfowl infected by AIV don’t show clinical symptoms. But since2002, H5N1highly pathogenic avian influenza virus can cause high lethality. In April2004, the epidemic of H5N1HPAFV infecting wild bird firstly outbroke in Qinghai Lake in China and the virus spread to other European and Asian countries in the following years, which reveals that wild bird plays an important role in the spread of avian influenza virus.There is a substantial production of domesticate ducks in China. Despite the side effects of H7N9, the annual output of domesticate ducks reached3.9billion. Different serotypes of avian influenza virus can be isolated from intestinal tracts and faeces of apparently healthy ducks in live poultry markets, and most of these viruses are highly pathogenic to chickens. Many serotypes were firstly isolated from ducks and then can be detected from birds, pigs as well as human, which indicates that domestic ducks has a critical role in the evolution and spread of avian influenza virus. With the development of molecular biology, many key amino acids related to virulence have been identified. Less studies have been made on the molecular mechanism that why H5N1HPAIV and LPAIV have different pathogenicity after infecting domestic ducks and the changes of different protein expression are poorly understood.In this study, we selected two H5N1AIV, R-GS/65and GS/HB65-PA224P+383D with highly genie homology and different pathogenicity, to infect domestic ducks and study the changes of host proteomics. As these two viruses cause different neural symptoms, we chose brain as the target tissue. We collected their brains after3days and4days, and the hypersensitive2-DE DIGE and Mass Spectrum were used to analyze the proteomics changes of brains. Meanwhile, we also used RT-PCR and Western Blot to further study some of the different expressed proteins. The result showed that fifteen and sixteen different expressed proteins were identified after3days and4days. Most of these different expressed proteins were involved in energy metabolism and nervous regulation. The study laid a theoretical foundation to further illuminate the pathogenesis of waterfowl infected H5N1AIV.
