ObjectiveBy detecting hypoxia-inducible factor1α (HIF-1α) and vascular endothelial growthfactor (VEGF) expression in maternal serum of patients with missed abortion and normalearly pregnancy and both expressed in the chorionic villi localization and expressionlevel differences, explore HIF-1α and VEGF role in the pathogenesis of missed abortionand both correlation analysis.MethodSelect the patient in obstetric of our hospital during2012.05-2012.10period andB-dynamic monitoring confirmed46cases of patients with missed abortion, which areknown possible causes of18cases (experimental group1), no clear reason for the28patients. Select the same period confirmed by the B-live births, and35cases ofpregnant women in our clinic line normal early pregnancy abortion as a control groupbecause of unwanted pregnancy.(experimental group2).Enzyme-linked immunosorbentassay was used to detect the concentration of HIF-1α and VEGF in serum andimmunohistochemistry was used to detect the localization and expression levels of thetwo cytokines in the experimental group and the control group villi.Result1.The expression of HIF-1α in serum and villus1.1The expression of HIF-1α in serum:①The serum concentrations of HIF-1α inthe experimental group and the experimental subgroup were lower than its concentrationsin the control group,and they have the statistical difference at the level of α=0.05.②HIF-1α in the experimental group1serum concentration less than its concentration in theserum of the experimental group, but there is no statistically significant differences (P>0.05).1.2Expression of HIF-1α in villus:①HIF-1α express both in the experimental and control groups villi, mainly expressed in the nuclei and cytoplasm of cytotrophoblastsand HIF-1α expression in the experimental groups were less than the control group, andthey have statistically significant differences (P <0.05).②HIF-1α in the experimental group1expression in the villous2villus expression slightly higher than the experimental group,but there is no statistically significant differences (P>0.05).2.The expression of VEGF in serum and villus2.1The expression of VEGF in serum:①VEGF concentration in the serum ofsubgroups of the experimental group and the experimental group were lower than itsexpression in the control group,and they have statistically significant differences (P <0.05).②VEGF in the experimental group1serum concentration comparing the experimentalgroup2serum concentration decreased, but there is no statistically significant differences (P>0.05).2.2Expression of VEGF in villus:①VEGF expression in the experimental andcontrol groups villus, mainly located in the cytoplasm and interstitial trophoblast, and theexpression of VEGF expression in the tissue of the experimental group obviously lessthan the control group,they have statistically significant differences (P <0.05).②Expressionof VEGF in the experimental group villi is slightly less than the expression in theexperimental group2villus, but there is no statistically significant differences (P>0.05).3.The correlation analysis of HIF-1α and VEGF3.1In serum:According to the Pearson correlation analysis, the expression ofHIF-1α and VEGF in the experimental group was significant with a positive correlation(r=0.578, P=0.008<0.01), the expression of HIF-1α and VEGF in the control groupshowed a positive correlation(r=0.549, P=0.012<0.05).3.2In villus:①The expression of HIF-1αand VEGF in the experimental groupvilli,Pearson relevant coefficient r=0.506, P=0.008<0.01was significant with apositive correlation.②The expression of HIF-1α and VEGF in the experimental groupvilli expression of PearsonThe correlation coefficient of r=0.352, P=0.021<0.05werepositively correlated. 3.3The correlation analysis of HIF-1α and VEGF in serum and villus3.3.1Missed abortion group (experimental group):①The expression of VEGF inserum and villus Pearson correlation coefficient r=0.436, P=0.030<0.05, showed apositive correlation between the expression.②The expression of HIF-1α in serum andvillus Pearson correlation coefficient r=0.50, P=0.035<0.05positive correlationexpression.③The VEGF expression in serum compare HIF-1α in the villi, Pearsoncorrelation analysis showed r=-0.187, P=0.370>0.05, no significant correlationbetween the two.④The expression of VEGF in the villus and HIF-1α expression inserum, Pearson correlation analysis showed that, r=0.390, P=0.110>0.05, respectively,without significant correlation.3.3.2Normal early pregnancy group (control group):①The expression of VEGF inserum and villus Pearson relevant coefficient r=0.324, P=0.038<0.05, showed apositive correlation between the expression.②The expression of HIF-1αserum and villusPearson correlation coefficient r=0.544, P=0.016<0.05, showed a positive correlationbetween expression.③The expression of VEGF in the serum and HIF-1α in the villi,Pearson correlation analysis showed that, r=0.300, P=0.057>0.05, respectively, withoutsignificant correlation.④VEGF expression in the villi and HIF-1α expression in serum,Pearson correlation analysis showed that, r=0.262, P=0.293>0.05, respectively,without significant correlation.Conclusion1.HIF-1α and VEGF expression may be low in the serum of patients with missedabortion is an important reason for angiogenesis during embryonic development, embryodevelopment stopped result in missed abortion.2.Tow expression of both HIF-1αand VEGF in patients with missed abortion villi isan important reason what may nourish blocked the causes Dysembryoplastic, causing thedeath of the embryo.3.HIF-1α in the serum of patients with missed abortion and villi low expression maycause the low expression of VEGF in serum and chorionic villi, the two were positively correlated.4.The expression level of VEGF and HIF-1αin missed abortion group and normalpregnancy group villi with both direct relationship and a positive correlation betweenserum content in missed abortion group and normal pregnancy group.5.In different body tissues, HIF-1α on VEGF may not achieve the regulatory role.
