Objective To develop a gene chip for purulen t meningitis common pathogensdetection,so as to provide new technology to confirm the diagnosis of purulent meningitisetiology.Methods Based on the design of specific probes for common pathogens of purulentmeningitis including Neisseria meningitidis bacteria, Streptococcus pneumoniae,Haemophilus influenzae bacteria, Staphylococcus aureus, Staphylococcus epidermidis,Escherichia coli and coagulase-negative staphylococci, we established a genechip assaysystem by optimizing the processes of chip substrate preparation, chip spotting andimmobilization, the sample amplification labeling and hybridization and chip scanning fordetecting the common pathogens of purulent meningitis. The developed genechip system wasused to detect the pathogen species of clinical samples for purulent meningitis diagnosis. Forclinical cerebrospinal fluid Samples were detected with and for sequencing verificationResults The results showed that among10CSF culturing positive clinical samples,8were detected by our developed biochip system as positive and2were negative. Another testshowed among36CSF culturing negative clinical samples,14were detected as positive bythe biochip system. The chip testing positive5cases verified by sequencing results with themicroarray consistentConclusions The genechip developed in this subject can be used for diagnosis andidentification of common pathogens of purulent meningitis. The positive rate wassignificantly higher than CSF culture. It is expected to be a high effective meningitis pathogendetection technology.
