Objective: To explore the difference of curative effect in myocardial infarction treated bytransplantation of Bone marrow Mesenchymal Stem Cell (BMSCs) between the diabeticand normal rats.Methods:The bone marrow were collected from80normal male SD rats of2months age,with20given high sugar and high fat diet (Additionally add lard and white granulatedsugar). The model of diabetic rats was established successfully, when the blood sugartaking from the rat tail>16.7mmol/L. The bone marrow of diabetic and normal rats werebeing adherent culture, and then the bone marrow mesenchymal stem cells were gottenunder aseptic condition. Application of flow cytometry to detect the second generation ofadherent cells. Drawing the Cell growth curve. The qualified cells were transplanted intothe focal area of myocardial infarction by direct injection. With successfully establishedmyocardial infarction model40normal rats were randomly divided into3groups(Group-1: group of normal rat with BMSCs transplantation, Group-2: group of diabetic rats with BMSCs transplantation and Group-3: sham-operated group). Aftertransplantation one month, Ejection fraction, the change of organization and the BCL-2were detected by cardiac ultrasound, HE staining and immunohistochemical methodrespectively.Results:Bone marrow mesenchymal stem cells were fusiform, arranged in spiralshape.Stem cell colony formation by of diabetic rats significantly smaller than the normalrats.Cells are unable to stretch and relatively flat.The capacity of cell proliferation isreduced.Flow cytometry instrument shows the expression of immune molecules on thesurface of the cells:CD90(96.12%);CD44(95.14%);CD29(94.51%);CD31(3.25%);CD34(1.17%);CD45(3.17%).the cell growth curve showed that normalrat bone marrow mesenchymal stem cells from diabetic rats bone marrow mesenchymalstem cells to grow fast.The study showed that the EF after transplantation1month in theGroup-1was significant higher than in the other two groups (P<0.05, respectively). InGroup-1, the myocardial infarction area organization form is better than that in Group-2,and also better than Group-3. There was no statistical difference between the Group-2andGroup-3about the cardiac function indexes (P>0.05).Conclusion:Through the observation of cell morphology and flow cytometry, the ratfemoral bone marrow adherent culture could obtain high purity of BMSCs. By means ofnormal and diabetic rats BMSCs transplantation for the treatment of myocardial infarctionconfirmed that diabetes reduced the curative effect of transplantation of BMSCs in treatingmyocardial infarction.
