Objective To compare carbon tetrachloride-induced liver cirrhosis in rats in twodifferent ways(intraperitoneal injection and intragastric), in order to build a lowmortality rate, high molding rate and good repeatability method. Method Fifty maleSD rats were randomized into3groups:In normal group A rats (n=10) wereintragastriced with pure peanut oil; In group B rats (n=20) were injected intoenterocoelia with the volume fraction of40%CCL4of peanut oil solution; In group Crats (n=20) were fed with the volume fraction of40%CCL4of peanut oil solution byintragastric. Three groups were fed according to the bodyweight changes based onthat of last week, twice a week,for a total of eight weeks. Three groups were on thenormal diet, with consumption of liquor mixed solution with distilled water (10%) ofethanol concentration, as drinking water, free to drink. Modeling process, the all ratswere consistently observed in terms of general condition, the activity and body weightchange. At the end of4th,6th,8th week of the experiment, the serum levels of alanineaminotransferase(ALT),aspartate aminotransferase(AST) of the rats were detectedand HE staining was performed on right liver tissues to observe the morphologicalchanges of the liver tissues. Result The rats in group A had normal activity, appetite,body weight gain and were formed normal flocculus. Rats in both of B and Cgroups had messy lackluster hair, poor appetite,mental fatigue, and slower responseto external stimuli.The survival rate of modeling made by CCL4intragastric(in groupC) was higher than that of modeling made by intraperitoneal injection.(in groupB).There was no significant difference in pathologic examination, ALT, AST betweengroup B and C.Conclusion Intragastric administration is better than intraperitoneal injection formaking a liver fibrosis animal model.
