0bjective:To study the expression of coagulant and fibrinolyticfactors in peripheral blood and placentas tissue of preeclampsia,analyzeits correlation with pregnant women and perinatal outcomes,so as toexplore the significance of balance disorders in the pathogenesis ofpreeclampsia patients.Methods:1. Plasma levels of t-PA、PAI-1were measured by ELISA in60casesof severe preeclampsia patients and60cases of healthy pregnant women。Plasma levels of PT、TT、APTT、FIB、AT-III、D-D、PLT、MPV were dedectedby auto coagulation analyzer STAGO STAR and auto blood cells analysissystem LH750.2. The expression levels of fibrinolytic factors t-PA、PAI-1in placentas of severe preeclampsia patients and healthy pregnantwomen were observed by immunohistochemistry.Results:1.Compared with the normal pregnancy group,plasma levels of PT、TT、APTT、AT-III、PLT were all decreased while plasma levels of FIB、D-D、MPV increased in early onset and late onset preeclampsia patients.2.Compared with the normal pregnancy group,plasma levels of t-PA in earlyonset preeclampsia patients showed an decreasing tendency,while PAI-1wassignificantly increased(P<0.05);plasma levels of t-PA in late onsetpreeclampsia patients showed an increasing tendency,and PAI-1wassignificantly increased(P<0.05).3.Positive expression of t-PA、PAI-1were both observed in placentas of preeclampsia patients and normalpregnant women,and the expression levels in placentas has signicantcorrelations with the plasma concentrations。Compared with the normalpregnancy group,expression levels of t-PA in placentas of late onsetpreeclampsia patients showed an increasing tendency while PAI-1wassignificantly increased(P<0.05). Conclusions:1.In patients with severe preeclampsia,there is balancedisorders between coagulant and fibrinolytic factors,which influence thepregnant women and perinatal adverse outcomes;early onset preeclampsiapatients has serious hypercoagulability than late onset preeclampsiapatients,which maybe lead to higher incidence of neonatal complications.2.In placentas of patients with severe preeclampsia,the expression levelsof t-PA and PAI-1were abnormal,Which suggest that hypercoagulation stateobserved in preeclampsia may be placental origin.
