One shock is caused by a variety of causes of effective circulating blood volume, inadequate tissue perfusion, impaired cell metabolic disorder and pathological syndrome. Massive blood loss shock is caused by uncontrolled hemorrhagic shock, it is to belong to a kind of severe blood loss, surgical is one of the most common type of shock. Uncontrolled hemorrhagic shock because of massive bleeding or fluid loss, or large amounts of fluid accumulation in the third clearance, results in the decrease of effective circulating blood volume. Uncontrolled hemorrhagic shock in trauma caused by bleeding, esophagus varicosity burst.Early uncontrolled hemorrhagic shock fatality rate is high, it can lead to systemic inflammatory response syndrome (SIRS), and further can cause multiple organ dysfunction syndrome (MODS), and inflammatory factors in uncontrolled hemorrhagic shock in the process of development and its important role. Early uncontrolled hemorrhagic shock, inflammation tissue ischemia hypoxia and was going on, at this point, the body is stimulated to activation of inflammatory cells, which in turn produces a large number of inflammatory cytokines, including IL-1β、IL-6、IL-10and TNF-α in uncontrolled hemorrhagic shock the early inflammatory response plays an important role.Due to uncontrolled hemorrhagic shock, has become a common form of clinical severe diseases in recent years, more and more experts and scholars dedicated to the shock of the early control research, find the shenfu injection of shock early inflammatory factor has certain inhibitory effect, and inhibition of systemic inflammatory response, patients receive timely rescue and treatment for the precious time. This experiment mainly study the shenfu injection to the uncontrolled hemorrhagic shock early inflammatory factor whether it has a certain inhibitory effect, and then confirm the necessity of uncontrolled hemorrhagic shock patients early given the shenfu injection.Objective:By detecting the blank group, model control group and experimental group in the three groups of serum IL-1β、IL-6、IL-10and TNF-a change, validation the shenfu injection. For uncontrolled hemorrhagic shock in rats after early inflammatory factor has an adverse effect, and then inhibit the systemic inflammatory response. For uncontrolled hemorrhagic shock patient early application the shenfu injection a reliable basis for treatment.Methods:In animal feed room,we randomly selected thirty healthy Wistar rats, the body weight in220arrives250gram.Thirty healthy Wistar rats were randomly divided into three groups, each group of ten, respectively as the blank control group (group A, not build model of uncontrolled hemorrhagic shock, not inject the shenfu injection), the model of uncontrolled hemorrhagic shock group (group B, build model of uncontrolled hemorrhagic shock, not inject the shenfu injection)and uncontrolled hemorrhagic shock and the treatment group (group C, uncontrolled hemorrhagic shock model, inject the shenfu injection). Group A is not bleeding, blood vein before pushing the same measuring physiological saline injection. Group B building push after intravenous injection the same measuring physiological saline. Group C after building, according to push5mL/kg (weight) of intravenous inject the shenfu injection. Three groups of building successful and process from external carotid venous sampling2hours after the2.0mL, centrifugal collect serum after (2000r/min). Using enzyme-linked immunosorbent (ELISA) method in the three groups were separately measured serum IL-1β、IL-6、IL-10and TNF-α. Using the single factor analysis of variance (One-Way ANOVA), two two compared the LSD-t test, comparison of the differences between groups of data.Result:1. Group B and group C rats serum IL-1β,IL-6, IL-10and TNF-α were significantly higher than group A(P<0.01), uncontrolled hemorrhagic shock after the production of large Numbers of inflammatory factors, systemic inflammatory response is obvious.2. Group C and group B compared to rats, the serum IL-1β,IL-6and TNF-α concentration of the latter (P<0.01), but it is still far higher than group a in rat serum IL-1β, concentration of IL-6and TNF-a, namely the uncontrolled hemorrhagic shock after push the shenfu injection, significantly reduce inflammation factors.3. Group C compared with group B rats, IL-10levels higher than the latter (P <0.01),uncontrolled hemorrhagic shock, inflammation suppression factor (IL-10) concentration increased. The differences were statistically significant (P<0.01).Conclusion:1. Uncontrolled hemorrhagic shock after the production of inflammatory cytokines increased rapidly.2. And can effectively reduce the production of inflammatory markers after uncontrolled hemorrhagic shock.3. The shenfu injection can promote the generation of uncontrolled hemorrhagic shock after inflammation suppression factor increases. Above results indicate that, the shenfu injection to the uncontrolled hemorrhagic shock in rats showed inhibitory effect on the inflammatory factors.
