PARTⅠ EFFECT OF SHENFU INJECTION ONHEMODYNAMICS IN CANINE WITH CARDIOGENIC SHOCK Objective: To investigate the effect of shenfu injection on hemodynamics in canine with cardiogenic shock. Methods: Cardiogenic shock model of canine was established by ligating left anterior descending(LAD)of coronary artery.The canines with cardiogenic shock were randomly divided in glucose injection group, shenfu injection group,shenfu injection combined with dopamine group and dopamine(DA)group.The hemodynamics parameters were measured by multi-physiologic monitor.And the indexes of different groups were compared. Results: 1. The mean artery pressure(MAP),left ventricular systolic pressure(LVSP),ventricular pressure rise ratio during systolic period(+dp/dtmax),and ventricular pressure decay ratio during diastolic period(-dp/dtmax)decreased significantly following cardiogenic shock.Left ventricular end-diastolic pressure(LVEDP)increased.Heart rate(HR)was accelerated. 2. From 30 to 180 minutes of treatment, MAP, LVSP and ± dp/dtmax increased significantly in shenfu injection group.LVEDP decreased. HR has not changed. 3. From 30 to 180 minutes of treatment, MAP, LVSP and ± dp/dtmax increased significantly in shenfu injection combined with DA group.LVEDP decreased.HR has not changed. 4. From 30 to 180 minutes of treatment, MAP, LVSP and ± dp/dtmax increased shenfu injection; cardiogenic shock; dopamine; hemodynamics over-expression of TNF-α mRNA, IL-1β mRNA in myocardium were observed following cardiogenic shock. The over-expression of TNF-αmRNA, IL-1βmRNA were inhibited in shenfu injection and shenfu combined with DA group. Compared with glucose injection group, there was significant difference. The over-expression of TNF-α mRNA, IL-1β mRNA did not decrease in DA group. Conclusions: ET, TNF-α, IL-1β could be inhibited by shenfu injection. Myocardium damage relieved. The experiment showed shenfu injection probably decrease over-exprssion of TNF-α mRNA, IL-1β mRNA on transcription platform.
