BACKGROUNDEsophageal and gastric cancers are one of the most common malignant tumors in clinical practice, their causes are not clear to date. Tumor immunity is one of the important mechanisms by which the body monitors and inhibits tumor cells. Tumor infiltrating T lymphocytes play an important role in the local antitumor immunity.This study mainly uses immunohistochemical methods, detects the distribution of tumor infiltrating CD45RO+T lymphocytes in endoscopic biopsy tissues from esophageal and gastric cancers and probes into its relationship with pathological grading.OBJECTIVE1. Observe the distribution of tumor infiltrating CD45RO+T lymphocytes in endoscopic biopsy tissues from esophageal and gastric cancers.2. Compare the distribution and densities of tumor infiltrating CD45RO+T lymphocytes in endoscopic biopsy tissues from esophageal and gastric cancers of different pathological grading.METHODS1. Esophageal cancer tissues were divided into3groups:adenocarcinoma with II grading,15cases, squamous cell carcinoma with I grading,17cases, squamous cell carcinoma with Ⅱ grading,26cases. Gastric cancer tissues were divided into3groups:adenocarcinoma with I grading,12cases, adenocarcinoma with Ⅱ grading,17cases, adenocarcinoma with Ⅲgrading,15cases. Specimens were firstly fixed in paraformaldehyde, then dehydrated, embedded in paraffin, successively cut into4urn thick slices.2. Immunohistochemical SABC method was used, using mouse anti-human T cell CD45RO monoclonal antibody, to detect the distribution of CD45RO+T cells in each sample.3. The slices were firstly observed under optical microscope at low magnification, then the area with the maximum CD45RO positive cells was selected,5high power field was observed, the positive cells was counted, and the average number was calculated. The data was processed by SPSSS17.0statistical software, the differences between different groups were compared.RESULTS1. CD45RO positive T cells in esophageal adenocarcinoma, stained into brown in the cytoplasm and membrane, mainly distributed in stroma between cancer nests, in a cluster, flake or scattered, the distribution in the cancer nests was alike.2. CD45RO positive T cells in esophageal squamous carcinoma, stained also into brown in the cytoplasm and membrane, mainly distributed in stroma between cancer nests, in a cluster, flake or scattered, CD45RO positive T cells were also seen around cancer nests, within the nests were also found a large number of CD45RO positive T cells, but the density is lower than the stroma, occasionally scattered in germinal center.3. The number of CD45RO positive T cells was bigger in gastric carcinoma, stained also into brown in the cytoplasm and membrane, distributed in the nests and stroma with high density, in a cluster, flake or scattered, CD45RO positive T cells were seen around the gland pipe with an annular shape in well-differentiated adenocarcinoma.4. In the esophageal adenocarcinoma tissue with II grading, the average number of TIL was59.73±6.01, in the esophageal squamous carcinoma tissue with I grading, the average number of TIL was60.73±5.34, in the esophageal squamous carcinoma tissue with II grading, the average number of TIL was130.21±9.25. adenocarcinoma with Ⅱ grading vs. squamous carcinoma with I grading, the difference was not significant. adenocarcinoma with II grading and squamous carcinoma with I grading vs. squamous carcinoma with II grading, the difference was significant.5. In the gastric adenocarcinoma tissue with I grading, the average number of TIL was119.61±8.37, Ⅱ grading was145.36±7.54, III grading was185.31±6.39. The differences between each2groups among the3groups were significant.CONCLUSION1. Tumor infiltrating CD45RO+T lymphocytes were found in endoscopic biopsy tissues from esophageal and gastric cancers, mainly distributed in stroma, in the tumor cell nests with an uneven distribution, reflected the body’s immune response against tumor cells.2. CD45RO+T cells infiltration densities in esophageal cancer tissues were as such, adenocarcinoma with11grading vs. squamous carcinoma with Ⅰ grading, the difference was not significant. The density of squamous carcinoma with II grading was bigger than the densities of adenocarcinoma with Ⅱ grading and squamous carcinoma with Ⅰ grading, the differences wwere significant.3. CD45RO+T cells infiltration densities in gastric cancer tissues were as such, adenocarcinoma with Ⅲ grading> adenocarcinoma with II grading> adenocarcinoma with I grading.
