Objective:Through discussing the influencing factorsof C57BL/6mouse endometriosis model toestablish a C57BL/6endometriosis model by intraperitoneal injection. Improve the success rate of the endometriosis model in C57BL/6mice.Methods:Establish endometriosis model by intraperitoneal infection of endometrial tissues in C57BL/6mice, many influencing factors were discussed,in three stages:Stage OneObserved5palnted mice on day1,2,3,4,5,6,7,8,9,12,15,18and recorded the size, weight, shape of lesions.Stage TwoGoup A:20estrum accepted mice with10estrum donor mice.Group B:20accepted estrus mice with10donor estrum mice.Group C:20estrum accepted mice with10estrus donor miceGroup D:20estrus accepted mice with10estrus donor mice.Observed and measured the diameter of the each donor mice’s uterus before the planting, and the growth of the lesions after9days.Stage Three60cases of estrus mice both as accepter and donor were grouped by different planting numbers of endometrial tissue, each group contained10samples and platned by1,2,4,8,16,32pieces respectively, Observed the growth of the lesions after9days to see whether the planting numbers influenced the establishment of endometriosis model or not.Results:Stage One There was significant difference of the size of the lesions during day1to8(P<0.05) while no significant difference in the following days (P>0.05).There was significant difference of the total weight of lesions during day1to5(P<0.05) while no significant difference in the following days (P>0.05).Stage Two:Chi-square test was carried out between the group A and group B, P=0.020<0.05, made a significant difference in statistics.Chi-square test was carried out on the group C and group D, P=0.027<0.05, made a significant difference in statistics.Chi-square test was carried out on the group A and group C, P=0.020<0.05, made a significant difference in statistics.Chi-square test was carried out on the group B and group D, P=0.027<0.05, made a significant difference in statistics.Stage Three:The success rate of2palnted pieces(or less) group made a significant difference in statistics compared to32pieces(P<0.05) while there was no significant difference in other groups(P>0.05).The surviving rate of lesions and the number of planted pieces built a three-function relation:y=-10.034+18.314x-1.284x2+0.024x3,R=0.981; F=34.780,P=0.028<0.05; T-test of regression coefficients:t=-1.878,8.206,-6.671,5.830.P=0.043,0.015,0.022,0.028, P<0.05.Conclusion:Stage One1. Establishing endometriosis model by intraperitoneal infection of endometrial tissues in C57BL/6mice results a higher successfull rate as a simple,stable and reliable method.2. The weight of the lesions tends to be stable after5days and the size of the lesions tend to be stable after9days which suggests day9as the best observating time.3. The living lesions shows varities of appearance which is significantly associated with the planting time and the typical one should shape a transparent or translucent cyst with visible blood vessels surrounded.Stage Two The estrus cycle of mice truely influences of the establishment of the EMs model, so choosing the estrogen mice distinguished by vaginal cast-off smear can improve the rate.Stage ThreeThe surviving rate of lesions and the number of planted pieces built a three-function relation; The success rate of EMs shows no relevance with the number of planted pieces when the number exceeds four.
