Interferon Induction Protein4and Kind Of Rheumatic Arthritis Relevant Research

Objective: To explore the relationship between the interferon-inducible protein4(IFIT4) express level in peripheral blood mononuclear cells (PBMC) of rheumatoidarthritis (RA)patiet and specificity of RA disease,. Study on the clinical significance ofIFIT4in RA.Methods: Collected30cases of the1987American College of Rheumatology(American College of Rheumatology, ACR) criteria for the diagnosis of RA patientsand20healthy controls. The peripheral blood samples,To separate the PBMC from thistwo groups with the lymphocyte liquid,to separate and extract total RNA by Trizol,reverse transcriptase mRNA to cDNA;determine theIFIT4mRNA expression level inthe two groups by Real-time quantitative polymerase chain reaction (RT-qPCR) methodand compare their differences.Detect the IFIT4level with enzyme linkedimmunosorbent assay (ELISA) method in RA patients and healthy controls’peripheralblood serum.RA related inflammation factor’s determination: blood sedimentation (ESR)with Clostridium welchii, rheumatoid factor (RF), c-reactive protein (CRP) withautomatic immunization rate nephelometry method. Testing anti-cyclic citrullinatedpeptide (anti-CCP) antibody level with ELISA.Compare the correlation betweeninflammation factors and IFIT4express level and protein content of relevancy.Results: The IFIT4express level and protein level is higher in RApatients(9.70±2.41,5.23±1.08),than,the,healthy(6.23±1.58,1.46±0.39). The differenceof IFIT4express level and protein level between RA and normal control group havestatistical significance（t=6.51，t=2.54,P均<0.05）. There is no correlation betweenCytokine CRP, ESR, anti-CCP antibody, RF and IFIT express level and IFIT4proteinlevel in RAConclusion: The quantitative expression of interferon-inducible geneIFIT4mRNA and protein level is high in patients with RA, the expression difference have certain significance in diagnosis of patients with RA. At the same time,it hascertain value on the degree of disease activity and judgment of the severity. Whichestablish the basis for future study on its immunological function. in patients with RA...
