Objective To investigate the effect and mechanism of Anti-IL-5Polyclonalantibody in the treatment of allergic rhinitis.Methods0valbumin (OVA) absorbed to aluminum hydroxide was used toconstruct the allergic rhinitis model(group B),and the normal control group (group A),the Fluticasone propionate group (group C) and Anti-IL-5Polyclonal antibody group(group D) consisted of6rats for each. One spray of fluticasone propionate was givento group C once a day in each nasal cavity for7days.30μL of anti-IL-5Polyclonalantibody to group D once a day in each nasal cavity for7days. Group B rats weresacrificed. The percentage of eosinophils of collected the nasal lavage fluid and bonemarrow cell suspension were determined and the pathologic changes and expression ofGATA-3were observed.Result Reduction of eosinophils was seen in nasal lavage fluid and bonemarrow cell suspension of group D as comparing with group B(P <0.01;P <0.01). Ingroup B,there are changes of mucosal congestion and edema’s thickening mucosa withinflammatory cells infiltration mainly of eosinophils; in group D, the above mentionedchanges were much more ameliorated.Immunohistochemistry showed significantincrease of GATA-3expression in the nasal tissue of group B but much lesser than thatin group D. In group D, the average optical density values of nasal mucosaimmunohistochemistry was significantly lower than group B(P<0.01).Condusions Anti-IL-5antibody can sigificantly improve the symptom score ofallergic rhinitis in rat model. The mechanism involve in its ability of reduction ofeosinophil percentage of the nasal lavage fluid and bone marrow cellsuspension,reduction of the eosinophilic infiltration in nasal mucosa and the inhibitionof expression of GATA-3transcripition factor in nasal mucosa.In addition, Anti-IL-5 antibody can also significantly alleviate changes of nasal cilia sparse, nasalcongestion,edema and thickening of the mucous layer.
