Objective:Through the establishment of the model of imbalance of intestinal flora with allergic asthma, Observation the effects of neuropeptide on the rats with allergi c asthma in lung and intestine of IL-4,IFN-γ of T lymphocyte subsets of Thl/Th2, T-bet related transcription and Gata-3; to find empirical evidence on mechanism oftr eated Lung and Large Intestine simultaneously and correlation theory on viscera of" lung and large intestine".Methods:Based on SD male rats as the research object, establishing the model of i mbalance of intestinal flora by colonizing the gastrointestinal with Cefoperazone aque ous solution and Candida albicans; establishing the model of allergic asthma by intra peritoneal injection and atomization excitation with ovalbumin (OVA), aluminum hyd roxide (Al (OH) 3), "jia-wei-sheng-jiang-san"is the representative prescription of tre ating Lung and Large Intestine simultaneously, through the therapeutic intervention with traditional Chinese medicine compound, take the research ideas of synchronous observation on Lung and intestine, observating the pathological changes of Lung and intestine with the method of Pathology, calculating the content of IL-4, IFN-γ of serum and bronchoalveolar lavage fluid (BALF) with ELISA, testing the content of the transcription factor T-bet, Gata-3 of Lung and intestine with immunohistochemic al method.Resuts:The lung index in the model group was significantly increased than the contr ol group (p<0.05),Contrary,Spleen index was significantly reduced (p<0.05). The lung index in the treatment group was significantly decreased than the model group (p <0.05),Contrary,The lung index was significantly increased (p<0.05);BS and LBS in m odel group was significantly reduced than the control group (p<0.05),The EC and The EMB and The Candida albicans in model group was significantly increasedthan the control group (p<0.05).Probiotics increased and Harmful intestinal bacteria redu-ced after treatment (p<0.05);The IL-4 in serum and BALF in model group was signi -ficantly increased than the control group (p<0.05),The IFN-γ in serum and BALF in model group was significantly attenuated than the control group (p<0.05). The IL-4 in serum and BALF in the treatment group significantly less than the model group (p<0.05),The IFN-γ in serum and BALF in the treatment group was significantly increased than that in the model group (p<0.05); The positive signal of T-bet was weakened in the model group than the control group (p<0.05),In contrast, Gata-3 was enhanced in signal (p<0.05).The Tbet positive signals increased in the treatment groups than in model group (p<0.05),The positive signals of Gata-3 decreased in the treatment group than in model group (p<0.05)Conclusion:One side, the expression of IL-4 can be down regulated by neuropeptide, and the expression of IFN-γ can be up-regulated by neuropeptide, The balance of Th1/ Th2 by the regulation. On the other hand, the expression of Gata-3 in lung and intest ine tissue be suppressed by traditional Chinese Medicine, the expression of T-Bet in lu ng and intestine tissue was enhanced by traditional Chinese Medicine, the balance of T h1/Th2 by the regulation.
