| Objective: Using the TaqMan Real-Time Polymerase Chain Reaction to comparebetween the Scaling and Root Planing treatment(SRP) and the treatment withminocycline hydrochloride for Detection and Quantification of moderate to severechronic periodontitis subgingival plaque with Porphyromonas gingivalis (Pg). Observethe changes in Pg amount between two different treatments, and remarking the Pg levelsby following clinical indexes: Probing Depth (PD), Bleeding on Probing (BOP),Bleeding Index (BI), and correlation with the attachment level (AL). We would like tohave a good clinical outcomes and a better clinical guidance by performing the aboveresearch.Methods:50cases of moderate to severe periodonitis patients, who took thetreatment in the First Affiliated Hospital of Dalian Medical University Dental OPD,which among them23cases were male and the other27female, with mean age of43years old were selected for the purpose of the study. We selected a total of100teeth,one right and one left in each patient whose teeth met the following criteria, PD≧4mm,AL≧3mm. We divided the patient into two groups according to randomized controltrial design as non-drug therapy and drug therapy. For the non-drug therapy group, wegave Scaling and Root Planing. For the drug therapy group we gave Scaling and RootPlaning and periocline therapy. The clinical parameters including p Probing Depth (PD),Bleeding on Probing (BOP), Bleeding Index (BI), and correlation with the attachmentlevel (AL). were measured at baseline,1, and3months after treatment and subgingivalplaque samples were collected at the same time when the measurements were made.Thesubgingival plaque samples were subjected to TaqMan Real-time PCR for quantitative detection of Pg. Data of PDã€BIã€AL and constituent ratio of Pg was collected and OneWay ANOVA was used to analyze the data using SPSS11.5software. ThePositive rate of BOP was analysed by Chi-square test.The relation between level of Pgin subgingival plaque and clinical parametes such as PD BI,AL was analysed bycorrelation analysis (Correlation analysis method).Results:1.Real-time polymerase chain reaction (PCR) detection under drainplaque Pg, drug and non-drug treatment group are68.0%detection rate; After treatment,1month,3Initial treatment below before treatment.2.constituent ratio of Pg results show that constitute the two groups after treatment,1month,3standard below before treatment, the result was statistically significant (p <0.05); After each treatment group three standard less than1months after treatment, theresult was statistically significant (P>0.05); Drug treatment group after1month,3phase before for the treatment of the decrease of the amount more than not treatmentgroup, but the result was statistically significant (P>0.05).3.constituent ratio of Pg with the clinical index,such as PD, BI and AL intopositive correlation, result was statistically significant (p <0.05).Conclusion:1.Quantitative real-time PCR provides a reliable and valuablemethod for quantification of periodontopathogens in subgingival plaque samples.2.Periodontal disease which is treated by Scaling and Root Planing treatment andmedication will reduce the number of Pg in subgingival plaque,and can maintaina therapeutic effect in three months. Of the total,SRP Plays a critical role, adjuvantdrug may improvement of clinical treatment effect than treatment SRP only3.TheQuantitative of Pg in subgingival plaque before and after initial treatment and clinicalindicators PD,BI, AL show positive correlation. |
