Objective:To recombinate semenogelin I derived peptides and study the inhibiton activity and mechanism of human semenogelin I derived peptides to human sperm.Methods:Human Sg DNA and its N-terminal Sg and C-terminal Sg DNA were cloned into PET32a vector. Positive clonies were screened and tranforme into E.ColiBL21.Recombinant Sg and its peptides were induced and expressed in high competent E. coliBL21, and purified by Ni-NT A column. Inhibition activity assay was done by adding concentrations of semenogelin’s two peptides,200pg/sperm, t o human spermatozoa.Results:The peptide of Semenogelin that inhibits the activity of human spermatozoa.Both N-terminal and C-terminal Sg inhibit the activity of spermatozoa.Conclusion:The peptide of Semenogelin that inhibits the activity of human spermatozoa.Both C-teminal and N-terminal Sg is the inhibition peptide of the whole molecular Sg.And the inhibition of peptide is significant associated with time.
