Objective:To study the activity and mechanism of human sperm motility affected by human Semenogelin I derived recombinant peptide Sgâ… -52.Methods:Select motile sperm from normal semen through Percoll density gradient centrifugation, handling with or without demembranated/reactived treatment. Then add Sg 1-52 in both the two kinds of sperm models, use the computer-assisted sperm analyzer to observe the difference of sperm motility between them, reference to time and concentration, and then use the SPSS statistics to analyze. Finally, relate the difference of two kinds of sperm structure under electron microscope with motility consequence we observed, to discuss the mechanism of Sgâ… -52 affect sperm motility.Results:In the test without demembranated process, Sgâ… -52 with concentration 100pg/sperm could inhibit motility rate within 90min. It could inhibit motility rate, forward motility rate, ALH mean and STR rate when concentration increased to 200pg/sperm. Between different concentrations of sperm motility test, there were some differences present in the mean of VCL, VSL and BCF. In each test of concentration, date changed were time related.In the test with demembranated process, both the two concentrations of 100pg/sperm and 200pg/sperm could inhibit sperm motility with almost all the date decreased. Between different concentrations, differences were present in the mean of VCL and BCF, which did not as obvious as the test without demembranated. In each test of concentration, date changed were time related. Besides, test with demembranated has shown less concentration dependence. Conclusion:Sg 1-52 has evident sperm motility inhibitive activity and put on time /concentration dependence. This activity is stronger when it acting on demembranated /reactived sperm. Sperm cell membrane is important to sperm to keep its normal motility, to which Sg 1-52 is likely to respond, and then inhibit sperm motility. To those sperm with membrane damaged but with intact internal structure, Sg 1-52 is likely to respond to intra-cellular to inhibit their motility.
