BackgroundLeprosy is a chronic diseases which caused by the mycobacterium, skin and peripheral nerve are involve in Human beings are the main host of the mycobacterium and the source of infection. Untreated leprosy patients(mainly the multibacillary leprosy patients) can infect through the respiratory tract and skin. But under the same conditions of equal access to leprosy patients, only a few people suffered disease, this is because the leprosy patients with different degrees of cellular immune deficiency. The precise mechanisms of cells immunodeficiency is still unknown.Studies have shown that cells of different subsets of T helper cells (Th) dysfunction plays an important role in the pathogenesis of leprosy development. And Thl, Th2, Th17and Treg imbalance could lead to leprosy patients with immune dysfunction trigger and exacerbate the patient’s condition changes.Different type of leprosy performance is the result of numerous cytokines synergistic effects, Th cell subsets in the release of many cytokines play network adjustment effect in various stages of leprosy.ifferent cytokine plays a complex disease, the development and transfer process to promote and (or) inhibition.Moderating Effect of different subsets of Th cell cytokine network, may help us to clarify the incidence of leprosy development process. And lay the foundation for further study of the role played by the different Th cell subsets in the development of the incidence of leprosy pathogenesis.ObjectiveStudy of leprosy patients peripheral blood Thl, Th2and Th17cell-associated cytokines (IL-2, IL-4, IL-6, IL-10, TNF, IFN-γ and IL-17) expression level.Analysis of cytokines in the peripheral blood of patients in each type of leprosy patients imbalance, explore its role in the incidence and development of leprosy and possible pathogenesis.MethodsExperimental group of69patients, divided into cure group, are symptoms of group, are symptoms of new-onset group, leprosy reaction group, are symptoms of multi-bacteria group, are symptoms of less bacteria group, cure multi-bacteria group, cure less bacteria group. The healthy control group of20patients.Concentrations of IL-2、IL-4、IL-6、IL-10、 TNF、IFN-γ and IL-17in plasma were measured by Cytometric Bead Array (CBA).The data was analyzed by SPSS19.0.Results1、Concentrations of IL-2、IL-4、IL-6、IL-10、TNF、IFN-γ and IL-17in plasma of the cure group and the control group, the symptoms of group and the control group, the symptoms of new-onset group and the control group, the leprosy reaction group and the control group are significant differences (P<0.05).2、Concentrations of IL-10、IFN-γ and IL-17in plasma of the leprosy reaction group and the symptoms of group are significant differences (P<0.05).3、Concentrations of IL-4、IL-6and IL-10in plasma of the symptoms of new-onset group and the symptoms of group are significant differences(P<0.05).4、Concentrations of IL-2、IL-4、IL-6、IL-10、TNF、IFN-γ and IL-17in plasma of the symptoms of multi-bacteria group and the control group, the symptoms of the less bacteria group and the control group, the cure multi-bacteria group and the control group, the cure less bacteria group and the control group are significant differences (P <0.05).5、Concentrations of IL-10in plasma of the symptoms of the multi-bacteria group and the symptoms of the less bacteria group are significant differences (P <0.05).6、Concentrations of IL-2、IL-4、IL-6and IFN-y in plasma of the symptoms of multi-bacteria group and tthe cure multi-bacteria group are significant differences (P <0.05).7、Concentrations of IL-10、TNF、IL-17and IFN-y in plasma of the symptoms of less bacteria group and the cure less bacteria group are significant differences (P <0.05).Conclusion1、The Cytokines of IL-2、IL-4、IL-6、IL-10、TNF、IFN-γ and IL-17play an important role in the pathogenesis of the development of the leprosy.2、In the early stage of leprosy disease, the type of multi-bacteria patients immune mainly Th2reaction increases.3、The level of IFN-y, IL-10and IL-17may be related to the leprosy reaction, low levels of IFN-y, IL-10and IL-17may be more susceptible to leprosy reactions.4、 Patients of less bacteria group with immune response (Th1, Th2and Th17-type) is stronger than multi-bacteria group.5、The level of the IL-10and IL-17may be plays an important role in a protective immune response patients of less bacteria group.6、In the multibacillary type of leprosy patients Thl and Th2type immune responses are elevated. Less bacteria type of leprosy patients Thl, Th2and Th17immune responses were increased.7、After treatment in patients with the body to achieve a relatively balanced state.8、Have been cured of leprosy patients with clinical symptoms have completely disappeared, but the body’s immune function has not been restored to normal.
