Effection Of Fasudil On Th1/Th2 And Th17/Treg-related Cytokines In The Nasal Mucosa Of Mice With Allergic Rhinitis

Objective: This study investigated the function of Rho kinase inhibitor fasudil on the mRNA expression of IFN-?,Il-4,Il-17 and Foxp3 related to Th1,Th2,Th17 and Treg in the nasal mucosa of mice with ovalbumin-induced allergic rhinitis.At the same time,the therapeutic effect of fasudil on nasal symptoms,morphology and inflammation of nasal mucosa in mice with allergic rhinitis were observed.Method:the allergic rhinitis model of C57 mice was established by ovalbumin and aluminum hydroxide through intraperitoneal injection and nasal drip.C57 mice were randomly divided into three groups: an AR group,a fasudil group and a control group.The AR group mice were sensitized and challenged by ovalbumin(OVA)without treatment.The fasudil group mice were treated with fasudil using the same process of AR group.The control group mice were treated with normal saline.At first,the symptoms of allergic rhinitis were observed and compared in each group.Then,the nasal mucosa of each group was stained with HE,and the morphological structural of the respiratory mucosa of the three groups were compared.The mRNA expression levels of IFN-?,IL-4,IL-17 and Tregspecific nuclear transcription factor Foxp3 in the nasal mucosa of each group were detected using real-time fluorescence quantitative PCR.Result:the symptoms of allergic rhinitis in each group were compared.mild nasal scratching and sneezing were found in the control group,but no obvious runny nose was found.Compared with the control group,the AR group had obvious sneezing,scratching and runny nose(all P<0.05).The symptoms of allergic rhinitis in the fasudil group were relieved compared with AR group(all P<0.05).Histopathological comparison of the nasal mucosa stained with HE in the three groups.There was no mucosal edema with eosinophil infiltration in the control group.Compared with the control group,there was an increase in eosinophil infiltration(more than 10 eosinophils per high magnification)and mucosal edema in the nasal mucosa of the AR group.Compared with the AR group,there was a decrease in eosinophils(less than 10 eosinophils per high power mirror)in the fasudil group.Compared with control group,IL-4 and IL-17 were significantly higher in the AR group(P<0.05),while IFN-g and Foxp3 were significantly lower in the AR group(all P<0.05).Compared with the AR group,the fasudil group significantly decreased the levels of IL-4 and increased the levels of IFN-g ?Foxp3 and IL-17 with statistical significance(all P<0.05).Conclusion: Fasudil can inhibit allergic rhinitis symptoms and nasal mucosal inflammation in mice,and improves the imbalance of Th1/Th2 and Th17/Treg in the nasal mucosa.
