Objective To investigate the efficacy of BMP‐2and VEGF single and combined repair application for open bone defect with seawater immersion.Meathods60New Zealand rabbits were divided into A、B、C、D four groups randomly and averagely. Make1.5cm bone defect using the rabbits of bilateral radius and then put them into the seawater for three hours. Group A as control group, implant autologous iliac into Group A; Groups B、C and D as experimental groups, implant iliac、absorbable collagen sponge(ACS) and BMP‐2into group B; implant iliac、ACS and VEGF into group C; implant iliac、ACS、 BMP‐2and VEGF into group D. Animals were sacrificed and cut specimens in the1st、2nd、4th、8th、12th weeks after grafting, Each group took six specimens for each phase randomly and observe the healing after grafting for the open defect under seawater immersion by imaging observation、histologic observation、immunohistochemistry and environmental scanning electron microscope.Results (1) Every group rabbits X‐ray have no significant difference during1st and2nd weeks. During4tht、8h and12th weeks, Callus gray value of groups B、C and D compared with group A difference was statistically significant (P<0.05);The difference of callus gray value between groups B、C and group D was statistically significant (P<0.05); The callus gray value between groups B and C were no significant difference (P>0.05).(2)Average absorbance value of BMP‐2and VEGF no significant difference in four group between8and12weeks;Group A and Group B, VEGF average absorbance value difference was statistically significantn time point of1stnd、2and4th weeks (P <0.05), Group Cand Group BD are also; Group A and Group C, BMP‐2average absorbance value difference was statistically significantn time point of1st、2nd and4th weeks (P <0.05), Group B and Group D are also. The results of immunohistochemistry showed that BMP‐2and VEGF mutually promote in bone defect repairing early, and they combined application have a synergistic effect.Conclusion (1) Both of BMP‐2and VEGF can promote individually seawater immersion open bone defect repair.(2) BMP‐2and VEGF combined application can promote remarkably the seawater immersion open bone defect repair, and they combined application have synergetic effects.
