| Object: Choose the water extract of raw and fried fructus xanthii, compare the acuteand long-term toxic effects of the experimental animals. It will provide a scientificbasis for reasonable and security applications of clinic.Method: Acute toxicity study used mice to, observe and record its toxic effects anddeaths, make the routine blood and the pathological examination. Long-term toxicitystudy with rats’ gavage for12weeks, make examination and record the weight,routine blood, liver and kidney related biochemical indicators, and the pathologicalorganization.Result: Acute toxicity study: At the low concentration, there were no obvious acutetoxic reactions of mice. At the high concentration, raw and fried product were bothproduced varying degrees of acute toxic effects in mice, and the higher theconcentration, the stronger acute toxic reaction. At the same time, according to thedeaths of raw and fried product groups, acute toxic reaction and the pathologicaldamage degree, the toxicity of the raw product is stronger than the fried product.Long-term toxicity study: On the whole, long-term use of water extract from fructusxanthii had certain toxic effects on rats, but the toxic effects were mild and occurredvery slowly.12weeks of continuous gavage. Part hematology and biochemistryindicators of rats appeared a slight change until9th week. The liver and kidney organsslices of experimental animals showed that, long-trem gavage of raw and fried fructusxanthii had pathological damages on rats to a certain extent, and the raw groups weremore serious than the fried ones. The renal fibrosis indicators such as HA, LN, HPCIII showed that long-term gavege of raw and fried fructus xanthii had not causerenal fibros.Conclusion: According to the acute toxicity study, the raw product was more toxicthan the fried product. Although its acute toxicity had no completely eliminated afterprocessing, but to a certain extent, processing reduces the toxicity of fructus xanthii,to make it achieved the purpose of processing attennuated.Long-term gavege of raw and fried fructus xanthii: Part hematology and biochemistryindicators of rats appeared a slight change until9th week. But there were slightchanges, and it hadn’t appeared renal fibros.In conclusion, the raw and the fried fructus xanthii have no acute toxicity at themaximum doses in human body, so short-term use is safe. Long-term high doses use(human dose more than3times), there may be slight toxicity reaction after9weeks,mainly reflected in body weight, urea and cre had changed or been higher. Therefore,human take the raw and fried fructus xanthii more than9weeks in the clinic should beclosely observed, including body weight and blood biochemical indexes, liver andkidney organs.At the same time, the acute and long-term toxicity of fried fructus were significantlyweaker than the raw product. So we should be vigorously promoted the process offructus xanthii and its processed products in clinical application. |
