Objective:To detect the genotype and allele distribution characteristics of rs290487on TCF7L2andrs13266634on SLC30A8,compare the differences in gene polymorphism analysis of TCF7L2andSLC30A8between Kazak patients with type2diabetes and normal population,analysis the relationshipbetween this differences and the Xinjiang Kazak type2diabetes susceptibility.Methods:(1) Study subjects were consecutively collected from in-patients of endocrinology department,and healthy individuals were selected from health examination center,Clinical examination:data regardingmedical,social,and course of disease were obtained during an interrogation.Measured height,weight,bloodpressure,BMI,WHR,FPG,FINS,FCP,TG,TC,HDL-C,LDL-C.(2) Gene polymorphism of200samples,including100cases of Kazak patients with type2diabetes and100cases normal glucose tolerance,weredetermined by gene sequencing and polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP).(3)The data of two groups were analyzed statistically by SPSS17.0software.Results:(1) The CC, CT, TT genotype frequencies of TCF7L2rs290487loci in Xinjiang Kazakh T2DMgroup (39%,42%,19%) and normal glucose tolerance control group (20%,51%,29%) has statisticallysignificant difference (P<0.05);there was significant difference of C allele frequencies of distributionbetween T2DM group (60%) and normal glucose tolerance group (45.5%)(P<0.01), the risk ofsuffering from T2DM,C allele carriers are1.797times of the T allele carriers(OR=1.797,95%CI1.208to2.672).(2) rs290487sites of Kazakh type2diabetes group: FCP, FINS, HOMA-B of CC genotype wassignificantly lower than the TT genotype’s, and FPG of CC genotype was significantly higher than the TTgenotype’s,the difference was statistically significant (P<0.05or P<0.01),but the difference was notstatistically significant (P>0.05) between the CC and CT, CT and TT.(3) The CC, CT, TT genotypefrequencies of SLC30A8rs13266634loci in Xinjiang Kazakh T2DM group (23%、46%、31%) and normalglucose tolerance control group (27%、50%、23%) has no statistically significant difference (P>0.05);therewas no significant difference of C allele frequencies of distribution between T2DM group (46%) andnormal glucose tolerance group (52%)(P>0.05).Conclusions:(1) compared Xinjiang Kazak type2diabetic group with the normal control group, the geneC allele of rs290487loci is the Xinjiang Kazak type2diabetes risk allele;(2) rs290487loci mutation mayaffect β-cell function, leading to inadequate secretion of insulin to increase Kazakh T2DM prevalence risk;(3) the rsl3266634loci of SLC30A8gene were proved to be a strong correlation with the Chinese Hanpopulation T2DM,but this result suggests that this loci is not related with the Xinjiang Kazakh T2DMincidence.
