We studied the separation and extraction of juglone and its derivatives which was extracted by chloroform from Green Peel of Juglans Mandshurica Maxim(GPJM) ethanol extract. The chemical composition, chemical structure and relative content of volatile components in GPJM were analysed, and the antioxidant activity of ethanol extract from GPJM was also studied.The active substances were extracted with95%ethanol from GPJM and the ethanol extract of active substances was extracted with chloroform and ethyl acetate. We got the best eluent proportion to separate chloroform phase using column chromatography, thin layer chromatography and ultraviolet visible spectrophotometer. The composition and purity were determined using high performance liquid chromatography (HPLC), infrared absorption spectrum and nuclear magnetic resonance (NMR) method. The substance content of the second column chromatography11-1stream was95.5%. This substance was benzoquinones, containing hydroxyl group, carbon-carbon double bond and benzene ring. However, due to the material was too little, co ultimately determine its precise structure.Two methods of vacuum distillation and water vapor distillation were used to extract the volatile oil in GPJM, and gas chromatography-mass spectrometry (GC-MS) was used to analysed the structure and relative contents of the volatile oil chemical ingredients. Results show that the main volatile components extracted by vacuum distillation were:palmitic acid methyl ester, caryophyllene oxide, phytol,1,4-dimethoxy naphthalene and benzene methanol, b-eucalyptus leaf alcohol,1-(1,5-dimethyl-4-hexene)-4-toluene, y-GuYun epoxide,2-ethyl butyl acetate, butyl phthalate, nerolidol; and the main volatile components extracted by steam distillation were:palmitic acid methyl ester, dihydro kiwifruit lactone, pohytol, the horn ene,3-allyl-6-cresol, extraordinary fragrance tree epoxide, dibutyl phthalate and n-pentanoic acid.We studied the antioxidant activity of the ethanol extracts from GPJM by measuring the clearance rate of hydroxyl free radical and sodium nitrite, and optimized the extraction conditions (the solvent and extraction method). We also explored synergistic effect between the extract and vitamin C. The result shows that, the optimal extraction method is ultrasound, the best solvent was80%ethanol. Within certain concentration range, clearance of the sample and the sample concentration could be represented by polynomial. There was a certain degree of synergistic effect between extract and vitamin C, and the strength of the synergistic effect was related to the concentration of the extract.
