| Objective:Identification and analysis of the Ureaplasma urealyticum (Uu) serotype distribution of52cases of tubal infertility female reproductive tract.Methods:The infertile female patients who visited Run Run Shaw Hospital, College of Medicine, Zhejiang University due to the fallopian tubes unilateral obstruction, tubal bilateral obstruction or fallopian tubes imperforation were choose and ruled out other gynecological diseases (such as bacterial vaginosis, Chlamydia infections, fungal vaginitis, gonococcal vaginitis, trichomonas vaginitis). Their cervical secretions were collected and cultured on the Mycoplasma IST2Uu liquid medium.52cases of cervical secretions were Uu positive. Then, the Uu liquid medium of52positive cases were transferred to A7solid culture medium for two to five days and further confirmed by the typical colonies under the microscope. The Uu genomic DNAs were extracted from the Uu positive liquid culture medium for the template of Real-Time PCR.14various serotype-specific primers were designed for Real-Time PCR. The Uu serotype distribution of52cases of tubal infertility female reproductive tract was analyzed. Results:All the52Uu positive specimens cultured on the A7solid culture medium have the typical colonies of Uu under the microscope. The52cases of Uu positive specimens were amplified for the14kinds of serotype by Real-Time PCR. Results showed that single serotypes accounted for50%(serotype1accounted for26.92%, serotype6accounted for11.54%, serotype9and10accounted for3.85%equally, serotype3and11accounted for1.92%equally), mixed serotype accounted for21.15%and there are28.85%failed to genotyping.Conclusions:Serotype1during the serotypes of Uu isolated from52cases of tubal infertility female reproductive tract accounted the highest proportion, followed by Serotype6, mixed serotype also accounted a higher proportion. |
