Objective: Bauhinia (alias entada, martial law etc.), a genus Rumex VineAsparagus Bean (Bauhinia championii Benth.) dry rattan of Benth., is akind of herbal medicine in Guangxi ethnic characteristics, and also haswide distribution, abundant plant resources on the southeast coast of China.In order to provide experimental basis for further research anddevelopment of new drugs, this paper mainly studied the mechanisms ofBauhinia flavonoid on antioxidation, anti-inflammatory and analgesiceffects, and myocardial ischemia reperfusion injury.Methods:1. To investigate cleaning and depressant effects of Bauhinia championiiflavonoid(BCF)on free radicals by using the system of proceedingoxyradical (·O-2) and hydroxy radical (·OH) in vitro.2. The analgesic effect and anti-inflammatory action of BCF wereobserved by using the mice writhing test, the hot-plate test and the modelof mice ear swelling caused by xylol.3. To establish a rat myocardial ischemia/reperfusion injury model, whileBCF pretreatment with different doses was observed. Rats heart rate (HR),left ventricular end diastolic pressure (LVEDP), and left ventricular pressure maximum increase/decrease rate (±dp/dtmax) were determined.In addition, the content of CK-MB, SOD, MDA, Na+-K+-ATPase andCa2+-Mg2+-ATPasewere detected, areas of myocardial infarction werestudied by TTC method, and myocardial morphological changes wereobserved by HE staining.Results:1. Effects of BCF on scavenging free radicalsBCF at low, middle and high concentrations could scavenge·O-2and·OH(P<0.05vs. control). Meanwhile, it inhibited the production of·O-2.2. Effects of BCF on analgesic and anti-inflammatory effectsIn the analgesic test,compared with the control group, three doses of BCFwere significantly reduced the writhing responses induced by the aceticacid, and the maximum of inhibitory ratio was44.4%(P<0.01).3. Effects of BCF on myocardial ischemia reperfusion injury in ratsIn the I/R test, Compared with the control group, results showed that threedoses of BCF could markedly improve the mice’s pain threshold inducedby the hot-plate. In the anti-inflammatory test, high dose (1200mg/kg) andmiddle dose (600mg/kg) groups of BCF could significantly reducedswelling of mice ear, and the inhibitory ratio of high dose group was52.2%(P<0.01). Compared with model group, BCF decreased HR, LVEDP,MDA, and CK-MB, while reduced the area of myocardial infarction,increased±dp/dtmax, SOD and the activity of Na+-K+-ATPase and Ca2+-Mg2+-ATPase (P <0.05or P <0.01)with dose dependent. HE resultsindicated that BCF led to ameliorate pathological changes of myocardial tissue morphology.Conclusion: BCF has good effects on scavenging free radicals, analgesiaand anti-inflammatory, and significantly protective effect on MIRI rats.
