| Objective: With adjuvant arthritis as an RA animal model,polyarthritis index score,secondary swelling degree of the paw,histopathologic changes of the ankle joint were used to evaluate,which to observe the effects of total flavones from Bauhinia championii on adj uvant arthritis in rats.The levels of IL-1?,IL-6 and TNF-? were detected,which to explore preliminary the mechanism of total flavones from Bauhinia championii in AA rats.Methods: The model of adjuvant induced arthritis in rats was prepared by an intrader mal injection of 0.1ml complete Freund adjuvant into the foot pad of the left hind paw of ra ts.Animals which were induced successfully were randomly divided into model groups,tot al flavones from Bauhinia championii with low,medium and high dose group(1.7,3.4,6.8 mg/kg),tripterygium wilfordii polycoride tablet group(7.53 mg/kg).Rats were not injected as a control group.The body weight,systemic inflammation index score,polyarthritis index sco re,secondary swelling degree of the paw,immune organ index and histopathologic changes of the ankle joint were used to evaluate,which to observe the effects of different doses of t otal flavones from Bauhinia championii on adjuvant arthritis in rats.The levels of IL-1?,IL-6 and TNF-? were detected by ELISA,which to explore preliminary the mechanism of tot al flavones from Bauhinia championii in AA rats.Results:1.Effects of total flavones from Bauhinia championii on the weight of AA ratsThe body weight of the model group and the control group showed a gain trend of weight.The body weight of the model group rats was lower than that of the control group rats a nd there was a significant difference(P<0.01).These groups of total flavones from Bauhinia championii(3.4,6.8mg/kg)increased the weight of AA rat,which the body weight was rest ored and improved in different degrees.2.Effects of total flavones from Bauhinia championii on the global score of AA ratsIt not only caused the swelling of polyarthritis but also nasal congestion,swollen ears,tail nodules and other systemic immune response,when AA rats caused by inflammation.T he global score of AA rats peaked at d19~D22 and began to subside in d28.The total flavon es of Bauhinia championii in each group were able to lower the global score of AA rats,wh ich High-dose group of total flavones from Bauhinia championii(6.8 mg/kg)played a most significant role.3.Effects of total flavones from Bauhinia championii on polyarthritis index in AA ratsThe total flavones from Bauhinia championii can reduce the polyarthritis index score o f AA rats in each group,which the high dose group(3.4,6.8 mg/kg)significantly inhibited polyarthritis index of AA rats and had obvious significant difference compared with the con trol group(P<0.01).4.Effects of total flavones from Bauhinia championii on secondary paw swelling in A A ratsCompared with the control group,secondary joint swelling in the model group increas ed significantly and there is a statistical significance(P<0.01),which succeeded in establish ing a model of AA.The total flavones from Bauhinia championii in each group of secondar y paw swelling of AA rats were inhibited.The inhibition of high dose groups of total flavon es of Bauhinia championii(6.8 mg/kg)was more significant.5.Effects of total flavones from Bauhinia championii on spleen and thymus index in A A ratsCompared with the control group,model rats of spleen and thymus index increased sig nificantly and were apparently significant difference(P<0.01).Each treatment group had no significant effect on thymus index of AA rats.Only the high dose group had a significant in hibitory effect on the spleen index.6.Effects of total flavones from Bauhinia championii on Pathological changes in splee n of AA rats by HE stainingCompared with the control group rats,the structure of splenic corpuscle was abnormal,which the myeloproliferative white pulp extended to the red pulp and blurred with the red pulp.Red blood cells increased and inflammatory cell infiltration occurred in the red pulp.Total flavones of Bauhinia championii each group may improve pathological changes of spl een of AA rats to varying degrees.7.Effects of total flavones from Bauhinia championii on pathological changes in seco ndary ankle joints of AA rats by HE stainingCompared with the control group,the ankle joint of rats in the model group occurred s ynovial cells thickens,inflammatory cell infiltration,the formation of pannus,narrow joint cavity,destruction of cartilage and bone tissue.The total flavones of Bauhinia championii i n each group can make AA rat non-inflammatory side of the ankle synovial cell proliferatio n and reduce cell infiltration.8.Effects of total flavones from Bauhinia championii on IL-1?,IL-6 and TNF-? conte nts in serum in AA ratsCompared with the control group,the levels of IL-1?,IL-6 and TNF-? in the serum of the model group were increased.The results showed that the total flavones of Bauhinia cha mpionii in each group can reduce the levels of IL-1?,IL-6 and TNF-? in the serum to varyi ng degrees.Conclusion:(1)The total flavones of Bauhinia championii had a therapeutic effect on AA rats and the effect of high dose was more obvious.(2)The anti-inflammatory mechanis ms of treatment of total flavones of Bauhinia championii may be associated with inhibition s of IL-1 ?,IL-6 and TNF-?. |
