Objective:Bauhinia Championii belongs to the genus Bauhinia of the family Legumi-nosae.As the folk medicine of the Shes in China.It is bitter in taste and flat-nature,therefore,it has extensive pharmacological effects of dispelling wind and dampness,Qi and blood circula-tion etc.It can improve the symptoms of rheumatoid arthritis(RA),significantly reduce the pain and swelling of joints.The clinical effect is accurate.At present,the Pharmacophore basis of Bauhinia championii anti RA is not clearn In this paper,We focus on the Bauhinia Champi-onii and intended to the result of anti RA activity oriented,then systemic research on the chemical composition of this plant.The discovery of compounds,which possess significant anti RA activity,provide a scientific basis and foundation material for the development of effi-cient and low toxicity of anti RA new drugs.Furthermore,it lay the theoretical foundation of utilization of the medicinal plants of the Bauhinia Championii.Method:1.Smash the Bauhinia championii into coarse powder and extracted by 90%etha-nol,then extracts are got by extraction with petroluem ether and ethyl acetate,respectively.All the fractions were purified by means of silica gel column chromatography,Sephadex LH-20 and semi preparative HPLC separation method.Their structures were identified by physical and chemical properties and NMR,MS,IR,UV,X-Ray and CD spectroscopy technology,as well as comparisons with the data in literature.2.Isolation,culture and identification of rat prima-ry synovial cells.The anti RA activity was evaluated by MTT staining.Result:1.33 compounds were isolated from the extract from Bauhinia championii,which were identified as:5,7-dihydroxycoumarin(1),Umbelliferone(2),5,6,7,3',4',5'-hexanmethox-yflavone(3),5,7,3',4'-tetramethoxyflavone(4),5,7,3',4',5'-pentamethoxyflavone(5),3',4',5,7,8-pentamethoxyflavone(6),3',4'-methylenedioxy-5,6,7-trimethoxyflavone(7),apigenin(8),liquiritigenin(9),5,7,3',5'-tetrahydroxy-6-methylflavanone(10),3',',5,7-tetrahydroxyflavan-one(11),4',5-dihydroxy-7-methoxy-6-methylflavanone(12),dihydrokaempferol(13),3,5,7,4'-tetrahydroxy-6-methylflavanone(14),lupenone(15),Bauchampine A(16),?-sitosterol(17),stigmast-4-en-3-one(18),?-sitosterol palmitate(19),stearic acid(20),gallic acid(21),syringic acid(22),3,4,5-trimethoxyphenol(23),4-Hydroxy-3-methoxycinnamaldehyde(24),ethyl gallate(25),3,4-dimethoxyphenylethanol(26),1-(3',4'-Dimethoxyphenyl)-l-propanol(27),3-methoxy-4-hydroxyphenylethanol(28),rhusopolyphenol F(29),4'-O-methyldavidige-nin(30),2',4'-dihydroxy-4-methoxychalcone(31),Bauhichamine A(32),Bauhichamine B(33).2.The anti RA activity tests of the isolated compounds showed that compounds 2,7,21,29 and 33 can significantly reduce the survival rate of rat primary synovial cells in vitro.And their IC50 value is smaller.Conclusion:1.33 compounds were isolated from Bauhinia championii,including 15 Flavones and 18 non-flavone compounds.Among them,compounds 16,32 and 33 was a new natural compound.Except for compounds 3,4,5,7,17,21,22,23 and 30,the rest were all isolated from the Bauhinia championii for the first time.Compounds 1,2,6,10,12,13,14,16,17,24,26,27,28,29,32 and 33,these 16 compounds were all isolated from the genus Bauhinia for the first time.2.The compounds 2,7,21,29 and 33 have significant inhibitory effects on the proliferation of rat primary synovial cells.Suggesting that these compounds have good anti RA activity.
