Objective:Through in vivo experiments investigate Lily induced tumor cell apoptosis mechanism.Methods:1. Different concentrations of Lily polysaccharide process HepG-2cell. MTT method was applied to determine the anti proliferation effects, HE staining was used to observe the cell morphology changes, Annexin-V/PI double staining method for the determination of apoptosis rate.2. H22tumor cells were inoculated into mice, preparation of transplantation tumor, calculate tumor inhibition rate, thymus and spleen index; at the same time, apoptosis was detected with DNA agarose gel electrophoresis, expression of Western blot detection of Bax, Bcl-2, Caspase-3, Caspase-9protein.The Results:Lily polysaccharide on HepG-2has obviously of body outside anti-proliferation role, dang concentration for1mg/mL role time24h Shi, drug processing HE dyeing found cell volume variable small, nucleus deep dye; flow type cell operation determination cell is not plus drug processing early apoptosis died rate only for0.73%,5-fluoride urine pyrimidine early apoptosis died rate for20.15%, and lmg/mL, and2mg/mL of lily polysaccharide water extraction property early apoptosis died rate respectively for8.93%, and7.22%, early apoptotic rate decreases with increasing concentration has increased.Different dose of Lily polysaccharide water extraction property anti tumor rate are above35%, spleen index, and thymus index difference is unlikely to; DNA agar sugar coagulation rubber Electrophoresis law can observation to obviously of apoptosis died article with, and has must of dose dependence; Western blot law detection to Lily polysaccharide extraction property can raised Bax protein expression, and cut Bcl-2protein expression, activated Caspase-3, and Caspase-9of expression led to cell apoptosis died, to played anti-tumor role.Conclusion:Lily polysaccharide has significant in vitro antiproliferative effect of HepG-2, has obvious anti-tumor effect on H22tumor-bearing mice. Its mechanism may exert antitumor effects through induction of apoptosis, including the regulation of cell cycle, prevent its value; regulation of Bax and Bcl-2, activation of Caspase-3, Caspase-9, leading to cell apoptosis through mitochondrial pathway.
