Objective:To investigate the inhibited effects of tripeptide Tyroserleutide (YSL) on metastasis of Hepatocarcinoma to Lung, explore its possible mechanismMethods:1. Vaccinating human hepatocellular carcinoma cells by tail vein to set up a model of pulmonary metastasis in nude mouse to observe the inhibitory metastase effect of YSL from liver to lung.2. Transwell model was applied to find the optimal induced concentration of MHCC97and SK-HEP-1human liver cancer cells.3. Transwell model was applied for invasion of MHCC97and SK-HEP-1human liver cancer cells cultured in extract of lung by YSL in vitro.4. Gelatin zymography assay was applied for MMP-2and MMP-9activities from supernatant of MHCC97and SK-HEP-1human liver cancer cells cultured in extract of lung by YSL in vitro.5. Western blot was applied for matrix metalloproteinases (MMPs) MMP-2and MMP-9protein expression of MHCC97and SK-HEP-1human liver cancer cells cultured in extract of lung by YSL in vitro.6. Real-time PCR was applied for MMP-2and MMP-9mRNA level of MHCC97and SK-HEP-1human liver cancer cells cultured in extract of lung by YSL in vitro.Results:1. YSL could significantly inhibited the metastasis tumor numbers in lung from liver in a nude model.2. In the experiment of liver cancer invasion induced by extract of lung,the number of cells increased significantly with a certain concentration of the lung extract.3. YSL could significantly inhibited the invasion of human liver cancer cells cultured in extract of lung in vitro.4. YSL could significantly inhibited MMP-2and MMP-9activities from supernatant of human liver cancer cells by YSL in vitro.5. YSL could significantly inhibited MMP-2and MMP-9protein expression of human liver cancer cells by YSL in vitro.6. YSL could significantly inhibited MMP-2and MMP-9mRNA level of human liver cancer cells by YSL in vitro.Conclusion:The model of metastasis to lung in a nude mouse proof the anti-metastatic to lung from liver effects of YSL. The extract of lung could induce the liver cancer cells significantly. YSL could inhibit invasion of human liver cancer cells cultured in extract of lung, the anti-metastatic to lung effects of YSL is significant. YSL could down-regulated mRNA level and the activities and expression of protein expression to inhibit the metastasis.
