ObjectiveTo observe the effect of human Umbilical Cord Blood mesenchymal Stem Cells (hUCB-MSCs) transplantation on the1H-MRS parameters in the rabbits with cerebral ischemia-reperfusion.MethodsThe hUCB-MSCs abstracted from60~80ml of cord blood of full term babies were cultured in vitro. A total of40healthy adult rabbits were randomly divided into sham group (n=10), cerebral infarction (CI) group (n=10), stem cell group (hUCB-MSCs)(n=10), and saline group(n=10), the rabbits of the last three groups were established cerebral ischemia-reperfusion model with suture emboli method, stem cell group and saline group, with separately injection of saline and hUCB-MSCs2h after MCAO. On Id,14d after reperfusion, neurological deficits were evaluated by the neurological severity scores (NSS) in the last three groups. The volume of cerebral infarction and the level of NAA, Cho, Cr, Lac in center area of ischema were detected by using MR scanning. Complete blood cell count, serum biochemical parameters were also investigated at same time.ResultsThe neurological scores of hUCB-MSCs group significantly decreased, compared with CI group and saline group(P<0.05). There is no significant difference between CI group and saline group(P>0.05).There is no significant difference on the WBC、RBC、Hb、Plt、 AST、 Urea、Cr、TG、 Chol and Glu level between each group. Compared with Sham group, the hs-CRP level of CI group, saline group and hUCB-MSCs group decreased significantly(P<0.01). The results show that the hs-CRP level of hUCB-MSCs group is significantly lower than that of saline group and CI group (P <0.05). But there is no significant difference between CI group and saline group(P>0.05).Compared with CI group and saline group, the volume of cerebral infarction in hUCB-MSCs group decreased significantly (P<0.05).Compared with sham group, the ratio of NAA/Cr and Cho/Cr in CI group, hUCB-MSCs group and saline group decreased and Lac/Cr increased significantly (P<0.01). The ratio of NAA/Cr and Cho/Cr in hUCB-MSCs group was significantly higher (P<0.05), while the ratio of Lac/Cr significantly lower than than that of CI group and saline group (P<0.05). There is no significant difference in the ratio of NAA/Cr,Cho/Cr and Lac/Cr between CI group and saline group (P>0.05).Conclusion1. hUCB-MSCs treatment could improve the neurological function in rabbits with cerebral ischemia-reperfusion.2. hUCB-MSCs treatment could reduce the level of hs-CRP.3. hUCB-MSCs treatment could reduce the infarct volume.4. hUCB-MSCs treatment could increase the ratio of NAA/Cr、 Cho/Cr and decrease the ratio of Lac/Cr.
