This paper briefly states the principles of resonance light scattering technique and itsquantitative basis. Reviews and simple remarks are made on the applications of the techniquein the research of organic dyes regarding the aggregations, the interactions with biologicalmacromolecules, the effects on the conformation of interactions and the determination ofbiological macromolecules.There is no doubt of the role of proteins which are composed of various types ofnatural life in the most important material,behoove plays a vital role in the health of thehuman body.The main methods for the determination are ultraviolet absorption, fluorescencespectrophotometry and resonance light scattering techniques, et.This paper uses theResonance Rayleigh Scattering spectrum to analyze and detect the concentration of protein.Resonance Rayleigh Scattering method is a new quantitative analytical technique, because ofits simple operation, high sensitivity,which is widely applied to determination of biologicalmacromolecules, medicine etc.The RLS method has simple manipulation, and study on theresonance light scattering spectra, it has obvious advantages in the determination of biologicalmacromolecules, such as proteins and nucleic acids.Since the spectral analysis of protein, it is essential to have the study method andinstrument, In this paper, used the resonance light scattering method as the theoretical basis,in-depth studied on the determination of proteins by resonance light scattering method, anddesigned a protein detector for spectral analysis and the measurement of proteinconcentration.The following major innovative works were carried out:(l) In a flow injection device,uses sodium dodecyl sulfate as probes to do the RLSspectrum analysis of the protein content in human serum samples. the results’reproducibilityand stability were elevated.(2) Used a new probe of indigo carmine as the Resonance Rayleigh scattering probe forthe determination of protein content, improved the detection sensitivity,extended the scopeof testing.(3) The linear regression equation of the protein solution was obtained throughexperiments that draw the Resonance light scattering spectroscopy, and designed a proteinconcentration detection system according to the principle.
