Objective:1.The study of miRNA-25expression in different grade gliomas; to explore therelationship between its abnormal expression and malignant degree of gliomas;Studythe expression of DSC-2protein in gliomas and normal brain tissue, to determinewhether DSC-2is glioma related protein, and its relationship with glioma malignantprocess.3.Explore the relationship between miRNA-25and DSC-2, preliminary judgmentof miRNA-25in glioma whether regulation DSC-2gene expression.Methods:Collection of different pathological grades brain glioma specimens in29cases,including glioma grade Ⅰ in5, glioma grade Ⅱ in6, glioma grade Ⅲ in9, gliomagrade Ⅳ in9;normal brain tissue(control) specimens6cases, extracted miRNA andtotal protein; using fluorescence real-time quantitative PCR the detected miRNA-25,by western-blot and immunohistochemical detection DSC-2expression of glioma andnormal brain tissue samples.Results:1.miRNA-25expression in glioma: compared glioma I grade with the controlgroup showed low expression of miRNA-25, with the increase in glioma pathologicalgrade, miRNA-25according to the increasing trending; among normal group andgliama grade I, III, IV, glioma grade I and glioma grade III, IV, glioma grade II andglioma grade III, IV, glioma grade III and grade IV,which have significant difference(P <0.05); while between the normal group and glioma grade II, glioma grade Iand grade II showed no significant difference (P>0.05).2.Purpose the molecular weight of the DSC-2protein in the control group andthe glioma specimens were negative strips (95KD), the majority sample showabnormal strip (130KD), compared the relatively exception strip differences amongthe groups: normal group with a comparison between glioma grade III, IV, glioma Iand glioma grade III, IV, glioma grade II and glioma grade III, IV, glioma grade IIIand grade IV,which have significant difference (P <0.05); between the normal group and glioma grade I, II, glioma grade I and grade II, which showed nosignificant difference (P>0.05).3.The immunohistochemical found the DSC-2expression in the control groupand the tumor group, the analysis found: the normal group and glioma grade III,IV,glioma grade I and glioma grade III, IV, glioma grade II and glioma grade III, IV,glioma grade III and IV grade, which have significant difference (P <0.05); whilethe normal group and glioma grade I, glioma grade I and grade II, which showed nosignificant difference (P>0.05).The DSC-2And miRNA-25for correlation analysisfound:miRNA-25and DSC-2expression in glioma positive correlation (R=0.96, P<0.001).Conclusions:1.miRNA-25in different pathological grade gliomas abnormalexpression,glioma I grade compared with control group showed lowexpression,miRNA-25expression was increased accordingly with the increase in thegrade of Pathological trending.2. The DSC-2protein expression in human brain tissue and glioma tissues,with the increase in the grade of Pathological, DSC-2protein was increased graduallytrending.3. Analyzing prompted miRNA-25and DSC-2expression in glioma waspositive correlation (R=0.96, P <0.001).
