Objective: The aim of this thesis is to investigate the 4 miRNAs' differential expression between the normal human tissue. and neuroglioma tissue, namely miRNA-21,miRNA-106,miRNA-27, miRNA-124, and to detest these 4 miRNAs'different forms of expression in various levels of brain glioma patient's tissue. And it also analyzes the correlation between the severity of brain glioma and these 4 miRNAs. It provides a feasible method and a way of clinical diagnosis, grading,prognosis and genetic treatment study in the early brain glioma.Methods: We collected 46 postoperation glioma patients' fresh tissues samples in tumor area as the experimental group, and 20 young no-family-history-of-cancer cerebral trauma patients' brain tissues which were removed during the operation as the control group. According to glioma WHO, the experimental groups were classified into two groups: the low-level glioma group and the high-level glioma group. The low-level glioma group includes the first level and second level patients'samples. And the high-level glioma group refers to the third and fourth level patients samples. By analyzing the published gene expression chip database, we searched the expression quantity of miRNA-21, miRNA-106, miRNA-27, miRNA-124 in the tissues of human brain glioma patients. And we counted and analyzed the differential expression of miRNA-21 , miRNA-106 , miRNA-27 , miRNA-124 respectively between the experimental group and the control group, and between the low-level glioma group and the high-level glioma groupResults:1. There are expression differences between glioma patients's tissues and normal human brain tissues in the experiment of using miRNA-21,miRNA-106, miRNA-27,miRNA-124.2. Glioma patients' MiRNA-21, miRNA-106, miRNA-27 expression quantity are higher than the healthy people, but their expression quantity of miRNA-124 is lower than the healthy people.3. Compared the expression quantity of MiRNA-21 in the tissues of high-level Glioma patients and low-level patients with the expression quantity of MiRNA-21 in the tissues of healthy people, there is a statistically significant difference comparison among any two groups, P<0.05).4. There is statistic difference in the miRNA-124 expression quantity of high-level glioma patients tissues and healthy people tissues (P<0.05). And the expression quantity of high-level glioma patients tissues is lower than those of healthy people tissues. In addition, there is no significant difference between the expression quantity of high-level glioma patients tissues and those of the low- level glioma patients tissues (P>0.05).Conclusion:1. MiR.NA-21 , miRNA-106, miRNA-27 and miRNA-124 are expressed in the tissues of glioma patients, which suggests that those four factors may be involved in the formation of glioma.2. MiRNA-21 is expressed differentially among the tissues of the high-level gliomas patients, the low-level glioma patients and the healthy human brain glioma patients. With the increase of the severity of glioma, it tends to rise. And this tendency reveals that MiRNA-21 may take part in the formation, development and the process of deterioration.3. The expression quantity of miRNA-124 is low in the high-level glioma patients tissues, which tells us that miRNA-124 may be effected as a inhibition cancer gene during the formation, development and the deterioration process of high-level glioma.4. MiRNA-21?miRNA-106?miRNA-27?miRNA-124 detested in the tissues provides reference value for the noninvasive diagnosis of early glioma, clarification and prognosis.5. MiRNA-21?miRNA-106?miRNA-27?miRNA-124 detested in the tissues offers the possible targeted spots in the study of gene treatment for glioma.
