Objective:To investigated the correlation among miRNA-21, miRNA-221、miRNA-222and the occurrence and development of glioma by detecting the expression of miRNA-21, miRNA-221、miRNA-222in the blood of glioma patients. It provided a feasible method and a way of early diagnosis and progression of judgment in the glioma.Methods:We confirmed60glioma patients through their pathological results, with15healthy samples as the control group. The experimental group were classified as low grade glioma group, the III level and IV level according to WHO grade, grade I and grade II of glioma. MiRNA-21, miRNA-221, miRNA-222of glioma patients and healthy people were detected in the blood by RT-PCR method. Then we analysed the data of miRNA-21, miRNA-221, and miRNA-222in the experimental group and the control group and the different level between high grade glioma and low grade glioma group. Results1.The expression of miRNA-21,miRNA-221, miRNA-222are higher than the healthy peole. 2.MiRNA-21had a statistically significant difference in expressionle vels in low grade glioma patients and high grade gliomas contrast to the control group (P<0.05).3. Compared with the normal population and low grade gliomas, miRNA-221’s levels in high-grade gliomas was statistically significant(P <0.05), but there was no statistically significant difference between the normal population and low grade gliomas(p>0.05).4.Theexpression levels ofmiRNA-222were significantly different on ly in the serum of high grade glioma groupand normal people (P<0.05), low grade glioma group and normal group and the group of high grade gliomas and low grade gliomas groups were small, not statistically significant (p>0.05). Conclusion1.The over expression of miRNA-21, miRNA-221, miRNA-222in the blood of glioma patients suggested that the three factors may be involve the formation of glioma.2.The expression of miRNA-21in the blood of the high grade gliomas group and low grade glioma patients was higher than the normal people group,which suggested that it could involve the formation, development, progression of glioma. 3. The high expression of miRNA-221,miRNA-222in the blood of the high grade glioma suggested that they may play an important role in the the formation and process of high grade glioma.4. There was no statistical significance difference of the expression of miRNA-222in the high grade glioma group and the low grade glioma patients, suggesting that the imbalance of miRNA-221, miRNA-222, miRNA-221may cause low grade glioma developing high grade gliomas.5.The detection of miRNA-21, miRNA-221and miRNA-222from the serum provided the reference for the early non-invasive diagnosis and the evaluation of glioma.
