Effect Study On Chinese Herbal Medicines To Intervent The Neuralgia Of Chronic Prostatitis

Objective: To observe the different effects of Chinese herbs among activating bloodcirculation, clearing heat antitoxicant and smoothing liver to regulate Qi on modeling ratsin order to explore the mechanism of Chinese herbal medicines to cure the pain of chronicnonbacterial prostatitis. Methods: In order to establish the experimental autoimmuneprostatitis(EAP)model, male Wistar rats were subcutaneously immunized with a salineextract of male rat prostate glands(RPG) in Freund’s complete adjuvant (FCA)andPertussis-Diphtheria-Tetanus vaccine0.5ml at the0thand30thday, the concentration was60mg/ml. The modeling rats were randomly divided into activating blood circulationgroup, clearing heat antitoxicant group and smoothing liver to regulate Qi group. All ofwhich had high, middle and low-dosage respectively. After a4-week medicineadministration the rats were sacrificed and the prostate tissues were taken for theobservation of histomerphological changes with the light microscope in all groups.TheIL-6, IL-8and PGE2levels of serum were detected by enzyme-linked immunosorbentassay(ELISA) and the tactile referred hyperalgesia was measured once a week using VonFrey Filaments since the beginning of the study. Results:(1)8weeks after treating withsaline extract of male rat prostate glands with concentration of60mg/ml.To some extent,the rats in EAP model group had suffered from chronic inflammation which includingweight loss, skin swell and fester. Comparing with control group, the degree oflymphocyte infiltration and interstitial proliferation showed significant difference in modelgroup (P<0.05).The result of ELISA showed that the IL-6, IL-8and PGE2levels of serumin model group were significantly higher than those in control group (P<0.05).Themechanical withdrawal threshold was significantly decreased along with the days afterexperiment (P<0.05).(2)After using the Chinese herb of clearing heat antitoxicant,compared with model group, the range of lesions had significantly reduced in all dosagegroups (P<0.05). The lymphocyte infiltration of prostate tissue had significantly reducedin low and high-dosage groups and the degree of interstitial proliferation in low-dosage group showed significantly reduced compared with model group. The result of ELISAshowed that the IL-6, IL-8and PGE2levels of serum in all dosage of clearing heatantitoxicant groups were significantly lower than those in model group (P<0.05). Themechanical withdrawal threshold was significantly increased along with the days aftermedicine administration (P<0.05).(3) After using the Chinese herb of activating bloodcirculation, the range of lesions and the degree of lymphocyte infiltration had significantlyreduced in all dosage groups compared with model group (P<0.05). The result of ELISAshowed that compared with model group the PGE2level of serum in high-dosage groupwas significantly lower (P<0.05). The IL-6level of serum in high, middle-dosage groupwere significantly lower (P<0.05). And the IL-8level of serum in all dosage groups weresignificantly lower as well (P<0.05). The trend of mechanical withdrawal threshold ofdifferent dosages intervention in different time point showed significantly differentbetween activating blood circulation group and model group (P<0.05).(4)28days afterintervening with the Chinese herb of smoothing liver to regulate Qi, the range of lesionsand the degree of lymphocyte infiltration had reduced significantly in all dosage groupscompared with model group (P<0.05). The result of ELISA showed that compared withmodel group the PGE2and IL-6levels of serum in high-dosage group were significantlylower (P<0.05). The IL-8level of serum in high, low-dosage group were significantlylower as well (P<0.05). The trend of mechanical withdrawal threshold of different dosagesintervention in different time point showed significantly different between smoothing liverto regulate Qi group and model group (P<0.05). Conclusion: Subcutaneously injectionwith a saline extract of RPG mixed with FCA and Pertussis-Diphtheria-Tetanus vaccinecan duplicate the EAP model successful.Low, middle dosage of clearing heat antitoxicant;high, middle dosage of activating blood circulation and high dosage of smoothing liver toregulate Qi are proved to reducing the prostatic inflammation and fibrosis obviously andcan protect the prostate tissue as well.
