Abstract/Summary:
Lots of research had shown the inflammatory factors played a key role in the development of atherosclerosis. Inflammation can occur at all stages of atherosclerosis and is also highly correlated with the accumulation of lipids in the arterial wall.This current genetic study of PLA2G7represents, to our knowledge, the first comprehensive htSNP analysis of PLA2G7to accurately capture the polymorphic variation of the gene inrelation to cardiovascular disease phenotypes.Lp-PLA2has emerged as a potential therapeutic target in coronary heart disease, AGT gene M235T variant has been proved in different racial and coronary heart disease.Whether Lp-PLA2gene R92H,V279F, A379V polymorphism and AGT gene M235T gene polymorphism is associated with Hubei Han population of coronary heart disease, In the present study,we investigated whether or not the interaction between the polymorphism and CAD risk factors have the ability to cause disease.To date,there are still no relevant studies have been reported at home and abroad.Objective:Analyze the distribution characteristics of R92H、V279F and A379V polymorphism loci of Lp-PLA2gene and M235T polymorphism loci of AGT gene in Hubei Han population, and related coronary heart disease with its risk factors, in order to investigate the pathogenesis of molecular genetics of coronary heart disease. Research the method of the detection of population genotype quickly and efficiently, screen susceptible gene of the population of coronary heart disease, check the genotype of the high-risk groups, take effective measures to reduce the risk of coronary heart disease, provide theoretical basis for the early diagnosis and prevention of coronary heart disease.Methods:One hundred and twenty-eight patients with coronary heart disease who were diagnosed by Coronary angiography in the first Affiliated Hospital of Yangtze University or have a history of myocardial infarction. The R92H,V279F.A379V polymorphism in the Lp-PLA2gene and M235T polymorphism in the AGT gene were detected by using Polymerase Chain Reaction-Restriction Fragment Length Polymorphism;Two-class non-conditional logistic regression analyse independent risk factors of coronary heart disease; SHEsis software for analysis of haplotype; Understanding of the interaction of R92H, V279F, A379V of Lp-PLA2gene and M235T of AGT gene polymorphism and risk factors of coronary heart disease.ResuIts:(1)The R92H site was detected GG,AG, AA of three genotypes. The V279F site was detected CC,AC of two genotypes, The A379V site was detected GG,AG,AA of three genotypes.(2) The genotype proportion of R92H and V279F in the coronary heart disease group and the control group has statistical significance(P<0.05). The genotype proportion of A379V in the coronary heart disease group and the control group has no statistical significance.(3) The R92H site and V279F site, R92H site and A379V site, V279F site and A379V site present linkage disequilibrium. The R92H site、V279F site and A379V site exist five type of haplotypes:A-A-G、A-C-G、 G-A-G、G-C-A and G-C-G.The distribution of A-A-G、G-C-G haplotypes in coronary heart disease and the control group has statistical significance.(4) The association analysis of lipid profile and genotype showed AC individuals in CAD group had significantly higher level of TG,compared with CC individuals.The AC and CC group have statistical significance.(5) The M235T site was detected CC,CT,TT of three genotypes, The genotype proportion of M235T in the coronary heart disease group and the control group has statistical significance(P<0.05).(6) The association analysis of lipid profile and genotype showed TT individuals in CAD group had significantly higher level of TG,compared with CC individuals.The TT and CC group have statistical significance.(7) In combined genotype analysis, the genotype of R92H、V279F of Lp-PLA2gene and M235T of AGT gene was significantly higher in patients with coronary heart disease than those without coronary heart disease (P<0.01).(8) By Logistic regression analysis of single factors and multiple factors for coronary heart disease found that coronary heart disease is a disease which was caused by genes and environment.age, HDL is an independent risk factor for coronary heart disease.Conclusions:The polymorphism of R92H and V279F of Lp-PLA2gene and M235T of AGT gene could be associated with risk of CHD in Han Hubei. Combined gene analysis of the V279F of Lp-PLA2gene and M235T of AGT gene may enhance the predictability of coronary heart disease.
