APS is extracted from the Astragalus polysaccharide,It is the most abundant astragalus,stronger immune activity of natural ingredients, studies have shown that astragaluspolysaccharide immune function and pharmacological effects, also has a wide variety ofsources, low cytotoxicity, enhance the immune effects, etc., can be used as a vaccine adjuvantfor experimental study.Candida albicans is widely sojourn in a host body condition of pathogenic fungi, present,for the treatment of Candida albicans infection is mainly through a number of anti-fungaldrugs, but systemic Candida albicans infection, there are still70%after taking antifungalmortality, so choose a good vaccine to stimulate the body’s adaptive and innate immunity willbe development of a safe and effective vaccine is an important strategy of Candida albicans.Study found that Candida albicans heat shock protein90(Hsp90) is an immune advantage ofCandida albicans antigen, is a major target for the immune response, can induce the body toproduce protective antibodies, of which C is a conserved epitope epitope, all patients withserum can react, so it can be used as a vaccine candidate antigen of Candida albicans.Expressed in our laboratory containing Candida albicans heat shock protein90epitoperecombinant protein C (rP-HSP90C), can be used as vaccines, recombinant vaccineimmunogenicity is poor but the need for a safe and effective adjuvant to enhance the immuneresults.In this thesis, astragalus polysaccharide (APS) as a vaccine adjuvant together withrP-HSP90C immunized ICR mice, mice were immunized subcutaneously twice after the lastimmunization the first14days, mice were immunized eye on blood, serum was collected forsubsequent experiments to study the APS immune enhancement. Immunized mice WesternBlot assay specificity of the antibody in the antiserum; ELISA assay specific antibody in theantiserum IgG, IgG1and IgG2b antibody titers; ELISA kit to detect the anti-serum IL-2,IL-12, IL-4, IL-10levels; delayed-type hypersensitivity test; kidney fungal load detection andanalysis of the pathological changes of renal biopsy studies APS on rP-HSP90C anti-systemicCandida albicans immune protection effect.Experimental results show that, APS and rP-HSP90C common immunized mice, specificantibodies in serum IgG, IgG1and IgG2b titers and IL-2, IL-12, IL-4, IL-10was significantlyhigher than rP-HSP90C group; enhanced delayed-type hypersensitivity; immune protectionexperiments showed, APS and rP-HSP90C common immunized mice cultured renal tissueCandida albicans colonies was significantly less than rP-HSP90C group by the kidneys losslevels decreased significantly. Experimental results proved favorable APS as rP-HSP90C vaccine adjuvant significantlyenhanced Th1-and Th2-type immune response, enhanced rP-HSP90C systemic anti-Candidaalbicans vaccine’s ability to APS in the field of vaccine adjuvants The application lays afoundation for Candida albicans infection vaccine experimental basis.
