The content of tannin from Beihua was analyzed by UV-chrominated hide powdermethod. The methodology of experiment results was investigated. The results showedthat the content of tannin from Beihua use UV-chrominated hide powder method isresemble with pharmacopeia method. The stability of the solution is well at20℃.After20minutes,tannin can be completly adsorbed by chrominated hide powder. The RSD ofprecision, repeatability is0.07%and0.64%respectively.The average recovery rat is98.86%,RSD is0.85%.The fatty acid and volatile oil of Beihua were studied by GC-MS.18peaks wereseparated,10compounds were identified from the fatty acids. Through data processing,the results show that the Beihua also contained abundant myristic acid and palmitic acid,they were37.50%and23.76%of fatty acids respectively. And50peaks were separatedand50compounds were identified from the volatile oil. Many kinds of chemicalcomponents were involved, such as ketone (27.65%), saturated group (2.58%), ether(18.22%), amine(4.33%), aldehyde (1.21%), alcohol (3.47%), carboxylic acid (23.50%).Using a ion-precipitation method, chose the Ca2+as the best precipitating agentfrom metal ions Al3+、Zn2+、Ca2+、Pb2+.the technological condition of separating tanninfrom Beihua was studied. The results showed that: The best treatment combinations inseparating tannin from the Beihua was: The Beihua in50mL extraction solution(solid-liquid ratio1:8) by adding Calcium hydroxide (1:16)8mL, reaction at30℃40min, and the extraction rate of tannin can reach20.74%by this treatment.Adsorption behavior of tannin from aqueous solution by the macroporouspolymeric resin D-101was studied through static experiments. The adsorption kineticswas studied based on the pseudo-first-order, pseudo-second-order and particle diffusionkinetics equations. Thermodynamic parameters for the adsorption were determinedusing the Van’t Hoff equation. Studies found that adsorption kinetics was described bypseudo-second-order kinetics equation. The mutual effects of boundary layer diffusionand intraparticle diffusion kinetics affected the adsorption processes of tannin on D-101resin. The empirical Langmuir equation was successfully employed to describe theadsorption process. The physical adsorption of tannin on resin was proposed. Theenthalpy value of adsorption of tannin on adsorbent was about5.5kJ/mol.The adsorption and desorption of tannin of Beihua with D-101macroporous resinwas studied. The results showed that when tannin concentration of the sample solutionwas1.04mg/mL,loading quantity of sample was20mL,PH was4,adsorption flow ratewas15mL/min,the adsorption rate is the best. When the adsorption flow rate was15mL/min the leak point was detected. The impact of elution agent and elution agentconcentration on elution rate was also being studied. The results showed that using 150mL70%ethanol, flow rate was15mL/min, can desorption the tannin completely.Concentrated liquid desorption was surrounded by vacuum drying. The content oftannin in the product≥90.8%.
