Tannins exists widely in about70%of the Chinese herbal medicine’sskin,branches, roots, leaves and fruits.It can also be called as the tannic acid.It has the haemostatic, antidiarrheal, convergence and multiple, these kindsof effect has been widely used to medical and life and other aspects. Withmodern pharmacological research level rise, Tannins’ various physiologicalactivity has been confirmed, as antiviral, bacteriostasis, antioxidant, mucousmembrane protection, antitumous effect and so on. Along with the research oftannins structure and chemical composition, it has been widely used in the oralclinical medicine. Persimmon in our plant area is a large and high output,since the ancient times is being regarded as medicine and food, Persimmonriches in composition, as polysaccharide, polyphenols, and the vitamin and soon,which showed significant antioxidant, detoxification, sterilization activity,etc.Dental caries and periodontal diseases is common oral diseases, whichmostly caused by infection of bacterium. In recent years, local use ofantibiotics are widely used in clinical treatment of oral disease, but thelong-term use of these broad spectrum antibiotics interrupted the oral normalflora’s balance, at the same time led to development of resistant strains anddestroyed the healthy oral micro-ecological environment. Natural plant hadsmall side effects to the human body, compared with the commonly usedmethods and anti-inflammatory drugs they have significant advantages. Sothrough the experimental research persimmon tannin water extract on theseoral common pathogenic bacteria inhibition of tannins compounds of dentalclinical reference has played an active role. Objective:Our persimmon resources are rich, at the same time, persimmon is rich intannins. It has the many kinds of physiological activity such as bacteriostasis,antiviral, antitumous effect, mucous membrane protection and so on. Researchof Tannins in stomatology has been developed step by step. The practicabilityand security of it in stomatology domain have been proved from manyaspects.The part of the persimmon tannin content determination andpurification process were less discussed, in order to promote the explorationand study of the tannin used in the oral treatment, further optimization ofpersimmon tannin purification process and determine the content of tannineach part has positive effect.With the development of oral preventive medicine, safe and effective,non-toxic side effects of oral diseases prevention and treatment has beenwidely attention. Currently there are still some shortages in the oral localprevention drugs. Some drugs are easy to cause oral bacteria group disorder.Tannins can effectively inhibit the growth of bacteria, safety and no side effect.It will soon be the oral and local drug new choice.Methods:1Persimmon fruit tannins extracted: Respectively to the persimmon fruit,persimmon, persimmon fruit peel for ultrasonic extraction,then obtainedPersimmon tannin Water extract after purification, whose molecular weight isabout10000. Adopt the Folin-Denis method to determine tannins content,and compare each part of the persimmon fruit content of tannin.2The persimmon fruit tannins water extract in vitro bacteriostasisexperiment: Streptococcus mutans, Streptococcus sanguis, Candida albicansand Escherichia coli were chosen as the experimental bacteria. They are all inthe form of freeze-dried preservation. The experimental strain cultured inColunbia blood plate medium or brain heart infusion agar medium. Theimproved liquid media two-fold dilution method was applied in thepersimmon tannin water extract in vitro were tested for antibacterial activity ofStreptococcus mutans, Streptococcus sanguis, Candida albicans andEscherichia coli. At the same time, set up two test-tubes that only contained nutrient broth and two test-tubes that only contained brain heart infusionrespectively as the positive control and the negative control. The experimentalbacteria to be used in the MIC test which is corrected the bacteriaconcentration by Densimat was added to the test solution test-tubes and thepositive control test-tube, eventually the bacteria concentration of eachtest-tube should became to5×104CFU/ml. The test-tubes were then incubatedat37℃for24hr.Then in a sterile environment in accordance with the originalmedium type were inoculated in the nutrient agar medium and brain heartinfusion agar medium, then incubated at37℃for48hr.Observed the lowestconcentration of persimmon tannin water extract which completely inhibitedof bacterial growth as the MIC.Results:1Tannins reference substance solution at0.0016mg/mL~0.0072mg/mL range is linearity, and the standard curve regression equation is y=39.91x+0.0141, correlation coefficient is R2=0.9984. Among them eachgram of the whole persimmon fruit contents396milligram of tannins, eachgram of the persimmon pulp contents297milligram of tannins, each gram ofthe persimmon peel contents59milligram of tannins.2Against Streptococcus mutans, MIC of the whole persimmon tanninwater extract was1:2, against Streptococcus sanguis, MIC of the wholepersimmon tannin water extract was1:4, against Candida albicans, MIC ofthe whole persimmon tannin water extract was1:32,against Escherichia coli,MIC of the whole persimmon tannin water extract was1:16.Conclusions:1Ultrasonic extraction method was used to extract tannin from the wholepersimmon fruit, persimmon pulp and persimmon peel, it is known from thecomparison that the whole persimmon fruit tannins content is the highest inother parts of persimmon fruit.Tannin content: the whole persimmon fruit> persimmon pulp> persim-mon peel.2The whole persimmon fruit tannin water extract in vitro to Streptoc- occus sanguis, Candida albicans and Escherichia coli showed obviousbacteriostatic activity, especially Candida albicans. The whole persimmonfurit tannin water extract to Streptococcus mutans also showed a certainbacteriostatic activity.
