The study in order to select more effective extraction method investigated threemethods (hot-water extraction, ultrasonic extraction, enzyme extraction) extractingpolysaccharides from Sedum aizoon L.. The methods of deproteinization, such asSevag method, TCA method, enzymic method and Sevag-enzyme connection methodwere also researched. The polysaccharides after deproteinization were isolated byDEAE-Cellulose and Sepharose G-200.The results were presented as follows:(1) Among these methods hot-water method has the highest polysaccharides yield,which is followed by enzymic technology. While the yield of ultrasonic method wasthe lowest. The optimized extraction conditions of hot-water technology wasextraction time95minutes, the ratio of materials to water1:14g/mL, extractiontemperature95℃, while the polysaccharides yield was (2.08±0.22)%. The optimalextraction process of ultrasonic method was extraction time23minutes, extractiontemperature52℃, extraction power128W. Under the conditions the actual yield ofpolysaccharides was (0.24±0.01)%. The optimization conditions of enzyme methodwas pH5.9, materials to ratio1:20g/mL, extraction time91minutes, and thepolysaccharides yield was (0.38±0.01)%.(2) The order of deproteinization was TCA method> Sevag-enzyme method>Sevag method> enzyme metod. The optimum conditions in Sevag field was time ofshaking25minutes, ratio of chloroform (CHCl3) and n-hutyl alcohol (C4H10O)3:1,the ratio of materials to Sevag regent2:1, repeated for4times. The protein removingratio was (61.28±0.25)%with polysaccharides loss rate (29.16±0.19)%under thisconditions. The protein removing rate and polysaccharides loss rate were(55.65±0.41)%and (35.72±0.02)%under the conditions as follows: reactiontemperature50℃, enzyme0.5%, reaction time90minutes, pH5.2. The conditions ofTCA method obtained by optimzing was TCA concentration10%, deproteinizationnumer4times, shaking for10minutes, under which the proteinization rate andpolysaccharides loss rate were (82.84±0.11)%and (32.64±0.08)%, respectively. Theproteinization rate and polysaccharides loss rate were74.99%and45.35%using Sevag-enzyme combined method, separately.(3) Three contents (SPS1, SPS2, SPS3) were obtained separating byDEAE-Cellulose column. Subsequently, SPS1was purified by Sepharose G-200obtaining two polysaccharide compents of SPS1-1, SPS1-2.
