| Schefflera arboricola Hayata belongs to Schefflera octophylla(Lour.)Harms andSchefflera venulosa (Wight et Arn.) Harms of Araliaceae. Its stem or leaf is ofscattering stasis pain and activating blood detumescence, and has been clinically usedfor treatment of injury, rheumatism, joint pain, gastric and duodenal ulcer pain, etc.The cure of existing preparations tablet for treating rheumatic arthralgia curative isremarkable. But it has a delivery problem for patients as it is the big dose ofmedication compliance. The previous report revealed that one of the effective parts ofSchefflera arboricola Hayata was organic acid. Therefore, in the present study, thetotal organic acid content and composition of fumaric acid were used as index withsupported by evaluation of drug efficiency in order to optimize purificationprocessing. Also the anti-inflammatory and analgesic activities of the products wereevaluated. All data taken together provide some useful information for upgradingpreparations, and a reference for the follow-up investigation on Schefflera arboricolaHayata.The content determination method of total organic acid and fumaric acid wasestablished first. Acid-base potentiometric titration method was used for measuringtotal organic acids content, and the high performance liquid chromatography wasapplied for determination of fumaric acid content. Then total organic acid and fumaricacid content were compared between reflux extraction technology, ultrasonic assistedextraction technology and microwave assisted extraction technology with differentsolvent, concentration, solid-liquid ratio, extraction time and extraction times, andthat experiment was designed by orthogonal experimental design. The sample thatcame from the best extraction was dealt with and optimized by alcohol sinkpurification technology. Therefore, the anti-inflammatory and analgesic effectbetween the original preparation and the extracts were compared by hot plate, aceticacid twisting, the ear swelling, changes in vascular permeability caused by acetic acidof animal experimental models. Results:The acid-base potentiometric titration method for determination of total organicacids content and the HPLC method for fumaric acid content were set up. The resultsof the present study showed that the best extraction process of Schefflera arboricolaHayata was water reflux extraction, with Liquid ratio1:15, extracting time15min,extraction times1, and the best alcohol sink process was one-time process of80%ethanol alcohol. Furthermore, that the product of water extract-alcohol precipitationhad a little more effect than that of the Schefflera arboricola Hayata tablet in theanimal experiments. |
