Schefflera octophylla (Lour.) Harms, which belongs to Schefflera of Araliaceae,shows various biological activities, such as anti-inflammatory and analgesic, expelwind-evil and remove wetness, anti-tumor and antiviral. Early research of our team hasshown that the ethanol extract and chloroform extraction had obvious analgesicanti-inflammatory effects, and found that the main compounds of the parts were pentacyclictriterpene and glycosides in S. octophylla.(1) The determination method of the content of total triterpene compounds wereestablished. With the oleanolic acid as the reference substance, the result showed that therewas a good linear relationship in the range of0.0832to0.28002mg, the regressionequation was Y=3.5335x-0.0559, r=0.9991.(2) Through single-factor experiment and orthogonal test, the optimal process were:water decoction extraction were used, material to solvent ratio was1:10, the extractiontimes were3times, every time was1h, then the concentration of extract was1.5g crudedrugs/mL and were precipitated by80%alcohol for8h, and took4BV extraction (1gcrude drugs/mL), then through the D101macroporous resin (the ratio of diameter and thehight was1:7) with1.0mL/min speed. With5BV of water and6BV30%alcohol to washthe impurities respectively, and collected the part by6BV of70%alcohol eluting. Thecontent and transfer rate of total triterpenoids of the enrichment part was61.91%and42.84%.(4) After giving drugs, the low and high dose of the enrichment part by macroporous resin could increase the pain threshold of mice significantly (P <0.01) after60,120minutes, reduce the number of acetic acid in mice body torsion significantly (P <0.01) and reduce the auricle swelling induced by xylene in mice rose signifi-cantly (P <0.05). Compared with the pharmacological activities of chloroform partof the prophase research, there was no significant difference (P>0.05). (5) At the montment, ten compounds had been isolated and purified and seven wereidentified by researchs:3-(4-Hydroxy-3-methoxyphenyl)-2-propenal(1), scheffurosideD(2),scheffoleoside D(3),3-epi-betulinic acid28-O-[α-L-rhamnopy-ranosyl(1â†'4)-O-β-D-glucopyranosyl(1â†'6)]-β-D-glucopyranoside(4),asiaticoside(5),3-O-β-D-xylopyranosyl-(1â†'2)-O-β-D-glucuronopyranosyl-29-hydroxyoleanolic acid-28-O-β-D-glucopyranoside(6),3-epi-betulinic acid-28-O-[α-L-rhamnopyranosyl(1â†'4)-O-β-D-glucopyranosyl(1â†'6)]-β-D-glucopyranoside(7).
