Context: Oxycontin, as one of the most familiar opinions, is used to provideanalgesia in cancer patients. In the clinical treatment, there are various effects on thepatients, which are due to the different quantities and functions of receptors,transporters and metabolizing enzymes that are influenced by their genetic codingpolymorphism.Objectives: This experiment preliminarily provide evidence of the distributionfrequency of the single nucleotide polymorphism (SNP) in OPRM1A118G, UGT2B7C802T, CYP3A4*1G, ABCB1C3435T and ABCB1G2677T/A in Chinese Hanpopulation and the connection between gene polymorphism and efficiency ofOxycontin.Methods: We collected data from57Chinese Han cancer patients (age range20-70years) who first received Oxycontin to ease pain. To compare curative effect before andafter treatment, they were divided into two groups (refectory group and remissiongroup) according to the degree of pain relief. Then PCR products which wereamplified from DNA of peripheral blood were directly sequenced. Finally, wecompared genotype and allele frequency between two groups using the chi-squaretest.Results:1. The variant G allele frequency of OPRM1A118G is48%, There was no differentgenotype distribution and different allele frequency between the two groups (P>0.05). 2. The variant T allele frequency of UGT2B7C802T is25%. Refectory group hasmore homozygous mutant (802TT) and less wild type (802CC) genotype than theremission group, which were statistical significance (P=0.047<0.05) but theheterozygote (802CT) is not statistically different between the two groups.3. The variant T allele frequency of CYP3A4*1G is27%. There was no differentgenotype distribution and different allele frequency between the two groups (P>0.05).4. The variant T allele frequency of ABCB1C3435T is37%. There was no differentgenotype distribution and different allele frequency between the two groups (P>0.05).5. The variant T/A allele frequency of ABCB1G2677T/A is52%. There was nodifferent genotype distribution and different allele frequency between the two groups(P>0.05).The frequencies of each genotype combination of ABCB1C3435T andABCB1G2677T/A did not show significant difference between the two groups usingthe reference haplotype (3435CC-2677GG).Conclusion: The patients’ curative effect of wild type (802CC) is better thanhomozygous mutant (802TT) at UGT2B7C802T. There are no association betweenthe single nucleotide polymorphisms (SNPs) and curative effect of Oxycontin incancer patients about OPRM1A118G, CYP3A4*1G, ABCB1C3435T and ABCB1G2677T/A respectively.
