The experiment show that malignant behaviors such as carcinogenesis and infectionare highly associated with the significant change of glycans. It is very important to reachthe structure of glycan.The glycans like antennas, transmitting information and signalbetween cell-cell.Results from recent studies showed the change of glycosyltransferasesexpression have the effects on tumor growth and metastasis.In laboratory studies, comparative the content of tetre-antennary branches wereincreasing in HCC and Pericarcinomatous Tissues. On this basis,we use HPLC toseparate and enrich the specificity glycan and use exoglycosidases for four glycans’sstructure analysis,respectively: NA2, NA2FB,NA3FBand high-mannose type glycanstructures.This paper had deeply research the relationship between GnT-IVa and tetre-antennaryglycans. In early-stage work found that increased GnT-IVa in HCC, this’s led tospeculation, GnT-IVa are associated with carcinogenesis. Examination of the expressionof GnT-IVa mRNA and protein in hepatocellular carcinoma cell line by qRT-PCR andwestron blot. Compared with the control group, BEL-7402GnT-IVa/1668group, theaverage inhibition rate of GnT-IVa was60%. On the basis, we found that the contentof tetre-antennary in BEL-7402GnT-IVa/1668group had down-regulated. Theexperiment result of MTT showed that the proliferation rate of BEL-7402and HepG2inGnT-IVa/1668group had down-regulated. In the wound healing test, compared theGnT-IVa/1668experimental group and GnT-IVa/NC group, in HepG2and BEL-7402cells, the healing rates have decreased, indicating the expression of GnT-IVadownregulation may make the ability of hepatoma cells’s movment become weaker.Compared with the control group, BEL-7402GnT-IVa/1668group after the number ofcells through Matrigel was significantly reduced, indicating that GnT-IVadownregulation causes liver cancer cell migration and invasion becoming weaker.In this paper, we use the exoglycosidase to analysis the structure of glycans. Theresults showed that we preliminary analysis four specific N-linked sugar chains.Glycosylation caused by GnT-IVa directs the content of three-antenna sugar chain,hepatocellular cell proliferation, movement capabilities, migration and invasion, hasdeclined. Thus, our findings establishes a certain foundation to deepen the study ofdeviation theory of preliminary analysis the structure of sugar chain structure as wellas GnT-IVa.
