Objective: To explore the effects of Combination of Puerariae and Salviae in qua-ils with atherosclerosis by feeding high fat diet, discuss its mechanism from impro-ving lipid metabolism, lipid peroxidation, the effective protection of the vascular end-othelial cells,compared with Simvastatin.Methods:The quail atherosclerosis model was reproduced by feeding high fatdiet(cholesterol1%,lard16%, propylthiouracil0.1%, Sodium cholate0.2%and basaldiet82.7%).The healthy quails were divided into7groups:the normal control group,the model group, the ethyl acetate extract of Puerariae and Salviae, the Water extractof Puerariae and Salviae and the Simvastatin group. Groups quail administered orallyonce daily for16weeks to observe the effect of Combination of Puerariae and Salviaeand Simvastatin on total cholesterol(TC),triglyceride(TG),high-density,lipoproteincholesterol(HDL-C),low-density lipoprotein cholesterol(LDL-C),Malondialdehyde(MDA),Super oxide dismutase(SOD), Endothelin(ET),Nitric Oxide(NO),Nitricoxide synthase(NOS),Oxidized low density lipoprotein(OX-LDL),ApolipoproteinA-iv(ApoAI),Apolipoprotein-B(ApoB)after16weeks. Meanwhile, calculate thecoefficient of quail liver and Photographs, and HE staining aortas organizations, at thesame time, to detect the pathological changes of the quail aortic tissue lipid plaque,endothelial and endometrial tissue.Results: Successfully copied quail atherosclerosis model with high fat diet for16weeks. Compared with the model group, the ethyl acetate extract and water extract b-oth could significantly lower serum TC, TG, LDL-C and VLDL-C levels, increasedHDL-C(P<0.05or P<0.01),reduce the atherosclerosis factor on TC/HDL-C, TG/HDL-C and LDL-C/HDL-C ratio (P<0.05or P<0.01)and significantly reduce theApoB, increase ApoA1,correct abnormal ApoA1/ApoB ratio (P<0.05or P<0.01); andsignificantly reduce the quality of the liver and liver quail factor (P<0.05or P<0.01);elevated serum NO content and NOS activity; reduce the content of ET-1and ET/NO ratio (P<0.05or P<0.01); lowering LDL and ox-LDL content (P<0.05or P<0.01));reduce quail serum MDA content, increased activity of SOD. Reduce foam cells anddeposition of lipid, improve swelling and degeneration of endothelial cell, suppressintimal thickening, and reduce the number of macrophages.(P<0.05or P<0.01).Conclusion: The Combination of Puerariae and Salviae can regulate the lipid meta-bolism, reduce the generation of lipid peroxides and cell adhesion molecule, improvemetabolic disorder of oxygen free radicals, enhance antioxidant capacity, Inhibit theformation of foam cells and plaque, protect vascular endothelial cells, which preventthe occurrence and development of atherosclerosis.
