Objective:Compared with Lovastatin,to observe the effeets of Curcumin on quails with atherosclerosis by feeding high fat diet.To discuss its mechanism from vaseular endothelium- protecting and suppressing vascular smooth muscle cell(VSMC)to proliferate.Methods:The model of atherosclerosis on quails was reproduced by feeding high fat diet(containing cholesterol 1%,bean oil 6%,lard 14%and basal stoyer 79%).Then healthy quails were divided into 4 groups:the control group,the model group,the Curcumin group,the lovastatin group.To observe the effect of Curcumin and Lovastatin on total cholesterol(TC),triglyceride(TG),low-density lipoprotein cholesterol(LDL-C), high-density lipoprotein cholesterol(HDL-C)after 2 weeks,4 weeks,6 weeks and 8 weeks. Meanwhile,HE staining and Weigert's staining were used to observe the pathology changes of the aorta of quails.Immunohistochemistry was used to detect the expression of proliferating cell nuclear antigen(PCNA)in the aorta of quails.Besides,to observe the rule of morbidity on quails with normal cultivation,quails were divided into 4 groups:age of 6 weeks,4 months,8months,13 months.To detect the change of TC,TG,LDL-C, HDL-C and the content of cholesterol in quails eggs from each group.And HE staining and Weigert's staining were used to observe the pathology changes of the aorta of quails.Results:The model of atherosclerosis in quails was successfully reproduced by feeding high fat diet(containing cholesterol 1%,bean oil 6%,lard 14%and basal stoyer 79%)after 8 weeks.Comparing with model group,the atherosclerotic plaque of aortic and the thickness of arterious endangium in Curcumin group were significantly decreased (P<0.05,P<0.01).The serum total cholesterol(TC),triglyceride(TG),low-density lipoprotein cholesterol(LDL-C)were obvious lower than those of model group(P<0.05, P<0.01).Compared with model group,high-density lipoprotein cholesterol(HDL-C)was higher than that of model group(P<0.05).Furthermore,Curcumin can reversed the enhanced expression of PCNA(P<0.01).In normal cultivation,compared with other 3 groups,TC,TG,LDL-C were higher in the group of 13 months old,and HDL-C was lower.Conclusion:Curcumin possessed significant anti-atherosclerotic action,which might be related to the adjustment of serum lipids level,protecting endangium,degrading the expression of PCNA and the inhibition of VSMCs proliferation.Though the research to quails under artificial breeding,it could be found that the level of atherosclerosis was more severer with the increase of the age of quails.
